Abstract: PURPOSE: An analysis method for identifying a material which regulates interaction between RNA molecule and protein is provided to simply and quickly identify the material and analyze specificity of sequence of identified compound. CONSTITUTION: An analysis for identifying an agent which controls interaction between ARE sequence fragment and target protein comprises: a step of selecting ARE sequence fragment and probing; a step of preparing a target protein which binds to the ARE sequence fragment and forms complex of ARE sequence fragment and the target protein; a step of detecting formation of the complex; and a step of selecting a candidate based on the complex formation. The ARE sequence fragment contains mRNA sequence of TNF-α, IL-1, IL-2, VEGF, c-myc, c-jun, N-myc, c-fos or Cox 2. The target protein is HuC protein.
Abstract translation: 目的:提供一种用于鉴定调节RNA分子和蛋白质之间的相互作用的材料的分析方法,以简单快速地鉴定材料并分析鉴定化合物序列的特异性。 构成:用于鉴定控制ARE序列片段和靶蛋白之间相互作用的试剂的分析包括:选择ARE序列片段和探测的步骤; 制备与ARE序列片段结合并形成ARE序列片段和靶蛋白质复合物的靶蛋白质的步骤; 检测复合物的形成的步骤; 以及基于复合形成来选择候选的步骤。 ARE序列片段含有TNF-α,IL-1,IL-2,VEGF,c-myc,c-jun,N-myc,c-fos或Cox2的mRNA序列。靶蛋白为HuC蛋白。
