Abstract:
A quantitatively analyzing method of melanin in a corneocyte for visually expressing distribution of melanin in the skin surface is provided to express melanin amount visually and quantitatively by quantification of melanin spot area per corneocyte, so that a user confirm the efficacy of skin whitening cosmetics easily. A quantitatively analyzing method of melanin in a corneocyte for visually expressing distribution of melanin in the skin surface comprises the steps of: (a) collecting a horny layer from the skin surface; (b) dyeing melanin in the collected horny layer; and (c) observing the dyed horny layer sample with a microscope, picturing the image of dyed horny layer sample, and quantifying the melanin spot area per corneocyte, wherein the melanin amount is classified into 5 grades; and a first grade corresponds to 0-5% melanin distribution per corneocyte, a second grade corresponds to 5-10% melanin distribution per corneocyte, a third grade corresponds to 10-20% melanin distribution per corneocyte, a fourth grade corresponds to 20-40% melanin distribution per corneocyte, and a fifth grade corresponds to more than 40% melanin distribution per corneocyte.
