본 발명은 서열번호: 1의 염기서열 중에서 선택된 활성 부위를 포함하는 프로모터; 상기 프로모터 및 리포터 유전자를 포함하는 벡터; 상기 벡터를 포함하는, 지방분해를 촉진하는 후보물질을 검색하기 위한 키트; 및 상기 벡터를 이용하여 지방분해를 촉진하는 후보물질을 검색하는 방법에 관한 것이다. 본 발명에서 제공되는 프로모터는 퍼록시좀 증식인자-활성화 수용체-감마(peroxisome proliferator-activated receptor-gamma, PPAR-γ)와 결합하여 멜라노코르틴 2 수용체 부속 단백질(melanocortin 2 receptor accessory protein, Mrap) 유전자의 발현을 증진시켜 부신피질자극 호르몬(Adrenocorticotropic hormone, ACTH)에 의한 지방 분해를 촉진하기 때문에, 상기 프로모터를 적절한 리포터 유전자와 융합시킨 벡터를 이용하면 지방 분해를 촉진하는 후보물질을 효과적으로 검색할 수 있으며, 이를 통해 얻어진 후보물질은 Mrap 유전자의 발현을 증진시켜 ACTH에 의한 지방 분해를 촉진하여, 비만 관련 질환을 치료 및 예방할 수 있다.

PURPOSE: A hair coloring composition is provided to improve hair coloring effect and color persistence, to reduce damage to hair, and to relieve scalp irritation. CONSTITUTION: A hair coloring composition contains one or more kinds of saponins extracted from Codonopsis pilosula Radix, red ginseng, Salvia miltiorrhiza Bunge, Scrophularia buergeriana, and Sophora flavescens Solander ex Aiton. The hair coloring composition contains 0.01-20 wt% of the saponins and has pH 6-12. The hair coloring composition improves color persistence or relieves damage to hair. The saponins form dense and rich pores.

A kit and a method for screening a candidate material which promotes the lipolysis is provided to increase the melanocortin 2 receptor accessory protein(Mrap) gene expression, promote the lipolysis by ACTH(adrenocorticotropic hormone) and prevent and treat the obesity relating disease. A kit for screening a candidate material which promotes the lipolysis comprises a vector containing a promoter and reporter gene. The promoter comprises 842th to 853th bases of the sequence number 1(SEQ ID NO:1) or additional 1569th to 1581th bases thereon. The reporter gene is a luciferase gene, enhanced green fluorescent protein(EGFP) gene, or beta galactosidase(lacZ) gene. A method for screening the candidate material which promotes the lipolysis comprises: a step of transfecting a test cell which is designated to express a PPAR-gamma(peroxisome proliferator-activated receptor-gamma) with the vector; a step of treating with the candidate material in the transfected test cell; and a step of measuring the expression degree of the reporter gene in the test cell and selecting the candidate material.
