Abstract:
본 발명은 월계수 알코올 추출물과 핵산 분획물은 인간 신경아세포종 SH-SY5Y cell을 OGD 처리한 인위적 뇌 허혈 모델에서 세포 사멸에 대한 보호 효과가 있는데, 월계수 클로로포름 분획물은, 인간 신경아세포종 SH-SY5Y cell의 뇌 허혈 모델에서, DAPK가 활성화 되는 탈인산화 과정을 억제함으로써 세포 자멸사로부터 세포를 보호하고, 조직 배양한 대뇌피질에서도 각 농도에 의존적으로 OGD 유도에 의한 hippocampus 세포 사멸 억제 효과를 있으며, 백서의 MCAO 모델에서도 효과적으로 경색의 범위를 감소를 확인할 수 있는 월계수 잎의 알코올 추출물과 클로로포름 분획물을 함유하는 허혈성 신경계 뇌질환의 예방 및 치료용 조성물에 관한 것이다.
Abstract:
PURPOSE: A composition containing alcohol extract and hexane fraction of Laurus nobilis L. leaves is provided to prevent cell apoptosis and to suppress dephosphorylation. CONSTITUTION: A composition for preventing and treating ischemic nervous system brain diseases contains alcohol extract and chloroform fraction of Laurus nobilis L. leaves. A method for preparing the alcohol extract and chloroform fraction comprises: a step of immersing dried Laurus nobilis L. in methanol at 40-50°C and filtering three times; a step of adding decompressing a part of methanol extract and adding 10% of hexane to the other part of methanol extract to separate to hexane layer and methanol layer; a step of decompressing hexane layer; a step of adding chloroform and distilled water(1:1, v/v) to the methanol layer; a step of shaking at 30-40°C two times to obtain chloroform layer and aqueous layer; and a step of fractioning aqueous fraction.

PURPOSE: A composition for preventing and treating neurodegenerative diseases and Parkinson's disease containing Laurus nobilis leaves is provided to suppress apoptosis and intracellular ROS(reactive oxygen species) and reduce overexpression of active caspase-3 and a-synuclein. CONSTITUTION: A composition for preventing and treating Parkinson's disease and neurodegenerative disease contains single ingredient isolated from hexane layer and chloroform layer of Laurus nobilis leaves. The single ingredient is costunolide, dehydrocostuslactone or spirafolide, or 3 alpha-acetoxyeudesma-1,4(15),11(13)-triene-12,6alpha-olide. The single ingredient isolated from chloroform layer of Laurus nobilis leaves is reynosin.
