Abstract:
PURPOSE: A nanofiber electro spinning scaffold is provided to control output rate of the virus vector and control the speed of gene delivery by controlling the content, temperature and the likes. CONSTITUTION: A nanofiber electro spinning scaffold which controls the output rate of adeno-associated virus vectors is manufactured by dispersing adeno-associated virus, AAV vectors on elastin-like polypeptide having amino acid sequence which is expressed by structure formula 1 and nanofiber electro spinning of ε-carprolactone. In the formula [VPGXG]n, X indicates amino acid except proline and n indicates 32-128. X is valine and n is 128. The number of the average molecular weight of the poly (ε -carprolactone) is 80,000. Abstract:
PURPOSE: A composite of magnetic nanoparticles and adeno-associated virus, and a gene delivery method into a cell using the same are provided to deliver a foreign gene into a non-invasive cell type, to control moving speed of a magnet, and to control foreign gene delivery speed. CONSTITUTION: A composite contains superparamagnetic nanoparticles which are conjugated through a streptavidin-biotin-NTA(nitrilotriacetic acid)-nickel-hexa histidine linker, and an adeno-associated virus(AAV) vector. The superparamagnetic nanoparticles are gamma-Fe2O3, Fe3O4, or a mixture thereof. The adeno-associated virus vector is AAV2 vector containing cytomegalovirus(CMV) promoter and hexa histidine at a 587th site of LQRGNLGHHHHHHSRQA-(corresponding to wild type AAV2:^583 LQRGNRQA). A gene delivery method into a cell comprises: a step of delivering a gene delivery system into the non-invasive cell type using the magnet; and a step of controlling moving speed of the magnet for controlling gene delivery speed.
