This invention provides a method of in vitro testing for evaluating a degree of stimulation imposed on an organism by an alien substance. The invention provides an in vitro stimulation testing method comprising such steps that, in aqueous solution, an alien substance (substance to be tested) is brought into contact with protein having absorption spectrum at its visible part, variation in absorption spectrum resulting from said contact is measured, and then an amount of said variation is evaluated as an index of stimulation on the organism caused by the substance to be tested. Further provided by this invention is a method of testing an organism stimulation by an alien substance which employs a combination of the abovesaid method with other in vitro stimulation testing methods. A kit for performing such testing methods is also provided. The methods of this invention show a satisfactory correlation with the Draze method as an in vivo testing method.
