Abstract:
본 발명은 5'말단에 티올기를 가지고 있는 선형핵산에 항암제를 물리적으로 결합시키고, 이후 금 나노입자, 압타머를 화학적으로 결합시킨 다기능성 핵산 기반 항암제, 이의 제조 방법 및 이를 포함하는 항암제 조성물에 관한 것이다. 본 발명에 따른 다기능성 핵산 기반 항암제는 A10 압타머의 사용으로 높은 표적지향성을 가지며, 고농축 항암제와 금 나노입자의 사용으로 열 치료/화학 치료의 이중 치료법(dual therapy)이 가능하여, 기존의 항암제에 비하여 낮은 부작용, 높은 항암 치료 효과 등을 가진다.
Abstract:
PURPOSE: A nucleic acid gel matrix for synthesizing nucleus mimic cell-free proteins, containing DNA, and translation and transcription factors, is provided to ensure similarity with natural genetic environment and to produce a large amount of proteins. CONSTITUTION: A method for preparing a nucleic acid gel matrix for synthesizing nucleus mimic cell-free proteins comprise: a step of depositing zinc oxide(ZnO) nanoparticles on a sapphire substrate and depositing gold on the surface thereof by an e-beam lithography or vacuum sputtering method; a step of treating the surface with 1 mM of an HDFT(heptadecafluoro-1-decanethiol) solution at room temperature for 15 minutes and preparing a ultra-hydrophobic surface; a step of binding single strand DNAs of sequence numbers 1, 2, 3, and 4 and preparing X-shaped nucleic acid nanostructure; a step of binding pIVEX plasmid and a DNA fragment using a restriction enzyme and T4 DNA ligase and preparing a recombinant plasmid; a step of binding X-shaped nucleic acid nanostrucrure, the recombinant plasmid, and transcription and translation factors using T4 DNA ligase and preparing a nucleic acid gel precursor; a step of placing the nucleic acid gel precursor on the ultra-hydrophobic surface, covering with PDMS template, and reacting at room temperature for 4 or more hours; and a step of reacting preparing nucleic acid matrix, a liposome prepared by membrane components(DOPC, DOTAP, Texas-red DHPE) at room temperature for 2 hours.
