Abstract: PURPOSE: A monoclonal antibody manufacturing which unites with the rtxa1antigen and a recombinant protein expression vector for offering diagnosis antigens are provided to facilitate mass expression and purification of the recombinant RtxA1(3491-4701). CONSTITUTION: A recombinant expression vector includes a sequence which codes 3491-4701 amino acids(the sequence number 6 (SEQ ID NO:6)) of the vibrio vulnificus RtxA1 antigen or a sequence which codes the fused protein. The recombinant expression vector is used for manufacturing the monoclonal antigen which unites with TrxA1 antigen or diagnosis antigen. A manufacturing method of the vibrio vulnificus RtxA1 recombinant protein comprises the following steps: manufacturing the recombinant expression vector; transforming the recombinant expression vector to a host cell; and producing the recombinant protein of the sequence number 6 or the fusion protein after cultivating the transformed host cell. The manufacturing method processes PCR by using a front primer represented by the sequence number 1 or by using a rear primer represented by the sequence number 2 before the first step.

Abstract translation: 目的：提供与rtxa1抗原结合的单克隆抗体制造和用于提供诊断抗原的重组蛋白表达载体，以促进重组RtxA1（3491-4701）的大量表达和纯化。 构成：重组表达载体包括编码突变弧菌RtxA1抗原的3491-4701个氨基酸（序列号6（SEQ ID NO：6））的序列或编码融合蛋白的序列。 重组表达载体用于制备与TrxA1抗原或诊断抗原结合的单克隆抗体。 创伤弧菌RtxA1重组蛋白的制造方法包括以下步骤：制备重组表达载体; 将重组表达载体转化至宿主细胞; 并在培养转化的宿主细胞后产生序列号6的重组蛋白或融合蛋白。 制造方法通过使用由序列号1表示的前引物或通过使用在第一步之前的序列号2表示的后引物进行PCR。

Abstract: PURPOSE: A monoclonal antibody produced from a hydridoma clone is provided to quickly and accurately diagnose, prevent and treat vibrio vulnificus sepsis. CONSTITUTION: A monoclonal antibody binds to vibrio vulnificus RtxA1 antigen prepared by hybridoma cells(deposit number KCLRF-BP-00261) and thermal-treated vibrio vulnificus RtxA1 antigen. The monoclonal antibody binds to 3981-4380th amino acid sites of vibrio vulnificus RtxA1. A composition for detecting vibrio vulnificus contains the monoclonal antibody. A diagnosing kit for detecting vibrio vulnificus contains the monoclonal antibody. A therapeutic agent for treating vibrio vulnificus contains the antibody.
