Abstract:
Assay devices are provided including a receptacle having a sample entry port; a plunger disposed within the receptacle; at least one reagent; a membrane attached to a first surface of the plunger; a wick providing capillary force; and a detection zone. The membrane includes a target conjugate zone in which affinity components are disposed. Capture compounds are immobilized in a detection zone, which is located between the target conjugate zone and the wick. A method of detecting a target analyte is also provided, including providing the assay device; providing a sample suspected to contain a target analyte; adding the sample onto the device; allowing the sample to travel along the membrane until the sample reaches the detection zone; immobilizing the target analyte through reaction of the target analyte with the capture compounds; reacting the immobilized target analyte with a reagent to generate a detectable signal; and detecting the generated signal.
Abstract:
A composite filter medium comprising a filter element comprising at least one porous fibrous filtration layer, and at least one layer of a sorbent, stationary phase particulates selected from organic or inorganic particulates having an average diameter of less than 50 micrometers, soft particulates, and ground monolithic particulates. The particulates are capable of binding target molecule by, for example, adsorption, ion exchange, hydrophobic binding, and affinity binding. The particulates provide higher binding capacities than can be achieved using filter media incorporating conventional process scale chromatography resin particulates.
Abstract:
A system for measuring diffusion of a compound across a membrane (190) generally comprises a membrane (190) held between a first base (150) having a plurality of outwardly extending hollow projections (155) and a second base (110) fastened to the first base and having a plurality of recessed tapered openings (115) therein adapted to engage the plurality of hollow projections. Methods for measuring membrane diffusion using such systems are included. The system components are also provided in kit form.
Abstract:
Controlled radical initiators, reaction mixtures containing the controlled radical initiators and various ethylenically unsaturated monomers, and polymeric materials formed from the reaction mixtures are provided. The controlled radical initiators are bis-dithiocarbamate or bis-dithiocarbonate compounds with a single carbon atom between the two dithiocarbamate or dithiocarbonate groups. Polymeric materials such as homopolymers, random copolymers, and block copolymers can be prepared using the controlled radical initiators.
Abstract:
A method of detecting a biological activity, comprising: providing, a sample that may comprise a first biological activity; a first indicator reagent that can be converted by the first biological activity to a first biological derivative; a gelling agent; and a plurality of particles, each particle comprising an organic polymer that receives and retains the first biological derivative, wherein the particles have a mean particle diameter between 0.05 microns and 20 microns; forming a hydrogel comprising the first indicator reagent and the plurality of particles; wherein at least a portion of the plurality of particles is dispersed in the hydrogel or disposed on a major surface of the hydrogel; bringing the sample into fluid communication with a portion of the hydrogel; and detecting the presence or absence of the first biological derivative, wherein the first biological derivative is retained by the plurality of particles; wherein detecting the presence or absence of the first biological derivative comprises detecting a fluorescent first biological derivative or a colored first biological derivative; and wherein the presence of the first biological derivative is indicative of the presence of a first biological activity.
