    Abstract: A method for isolating and culturing adult stem cells derived from human amniotic epithelium is provided to improve collection convenience as compared to therapeutic stem cells of umbilical cord blood and bone marrow, and increase yield and growth rates by DTT(dithiothreitol) treatment, ROCK(Rho-associated kinase) inhibitor addition or medium exchange. A method for isolating and culturing adult stem cells derived from human amniotic epithelium comprises the steps of: treating the human amniotic tissue with 5-50 mM of DTT; treating the DTT-treated human amniotic tissue with 0.025-0.125% of trypsin to isolate human amniotic epithelial cell; culturing the isolated human amniotic epithelial cell in DMEM(Dulbecco Modified Eagle Medium) or K-SFM(keratinocyte serum free medium) containing 10nM to 100muM of ROCK inhibitor; and recovering the stem cells from the cultured medium.
