Abstract: An isolated or purified nucleic acid molecule consisting essentially of a nucleotide sequence encoding a mutant human IGFBP-3, which can inhibit DNA synthesis, can induce apoptosis, binds to neither human insulin growth factor-I (IGF-I), nor human insulin growth factor-II (IGF-II), and comprises a mutation at Y57; a vector comprising the same, a cell comprising and expressing the same, optionally in the form of a vector; an isolated or purified polypeptide molecule consisting essentially of an amino acid sequence encoding a mutant human IGFBP-3, which can inhibit DNA synthesis, can induce apoptosis, binds to neither human IGF-I nor human IGF-II and comprises a mutation at Y57; a composition comprising the same; and a method of inducing apoptosis in a cell, which method comprises administering to the cell the nucleic acid molecule or polypeptide molecule, in an amount sufficient to induce apoptosis in the cell, whereupon apoptosis is induced in the cell.

Abstract: The invention provides a method of reducing cancer progression comprising administering a Mx polypeptide or Mx-encoding nucleic acid to a host, such that the growth rate of the cancer cells is reduced, the metastatic potential of the cancer cells is reduced, or both. The invention also provides a method of assessing the metastatic potential of a cancer comprising (a) obtaining a sample of the cancer, (b) determining the level of Mx, Mx-nucleic acid, or both in the sample, and (c) comparing the level of Mx, Mx-encoding nucleic acid, or both with a control. In another aspect, the invention provides a method of assessing the ability of an agent to modulate the level of expression of an Mx comprising obtaining a cell expressing a known level of an Mx; contacting the cell with an agent to be tested; and assaying the cell for expression of the Mx to assess the ability of the agent to modulate Mx expression; alternatively, the method includes contacting a cell comprising a stable nucleic acid comprising the MxA promoter or other MxA regulatory sequence operably linked to one or more reporter genes to identify molecules that operably target such MxA nucleic acid sequences.

Abstract: This invention relates to certain carbamate analogs of thiaphysovenine of the formula wherein R is H and R is C4 - C10 alkyl; R and R are independently C1 -C10 alkyl; or R is H and R is a group of the formula wherein R is H and R is an isopropyl group; including optical isomers of the 3aS series, racemic compounds of this type and processes for their preparation. Pharmaceutical compositions containing such compounds are also provided. Such compounds and compositions are useful as cholinesterase inhibitors, especially butyrylcholinesterase inhibitors.

Abstract: The present invention relates to novel glucocorticoid receptors that have greater affinity and specifity for glucocorticoid steroids than the naturally occurring receptors. More particularly, the invention relates to the altering of the equivalent of cysteine-656 of the rat glucocorticoid receptor to either serine or glycine for the production of super glucocorticoid receptors which retain full biological activity in intact cells and have higher affinity and specificity for glucocorticoid steroid binding than the original receptor. The invention further relates to the recombinant expression of such altered receptors in host cells.
