Abstract: PROBLEM TO BE SOLVED: To obtain the new subject DNA fragment containing a gene corresponding to biosynthesis of biotin derived from microorganisms belonging to the genus Sphingomonas, used for gene technological breeding of microorganisms producing the biotin useful as a food additive, etc. SOLUTION: This DNA fragment is a new DNA fragment containing genes corresponding to a biotin biosynthesis, such as a 7-keto-8-aminopelargonic acid synthase gene, a 7,8-diaminopelargonic acid aminotransferase gene, a desthiobiotin synthase gene, a biotin synthase gene and a gene of an enzyme catalyzing a reaction of a biosynthesis step upstream from Pimelyl Co-A in a biosynthesis rout of the biotin, and is useful for gene technological breeding, etc., of biotin-producing microorganisms. The DNA fragment is obtained by extracting chromosome DNA of Sphingomonas paucimobilis JCM7511, forming a chromosome DNA library by a conventional method, and recovering a DNA from strains capable of being cultured in a medium containing no biotin.

Abstract: PROBLEM TO BE SOLVED: To improve the optical purity of an amine compound used for an agent for optical resolution and pharmaceutical and agrochemical intermediates by making a culture product of a specific microorganism that preferentially metabolizes either one of optical isomers of a specific amine compound act on the amine compound. SOLUTION: This method for improving the optical purity of an amine compound of the formula [R is a (substituted) phenyl or a (substituted) 7-10C aralkyl; R is a 1-6C alkyl], which is useful as a basic agent for optical resolution, pharmaceutical and agrochemical intermediates, etc., comprises making a culture product of microorganism belonging to the genus Arthrobacter [for example, Arthrobacter SC-K99 (FERM-BP-5744) etc.] which has an ability to preferentially metabolize either one of optical isomers based on an asymmetric carbon atom combined with an amino group in the amine compound of the formula or its processed material act on an amine compound of the formula (for example, 1-phenylethylamine, etc.).

Abstract: PROBLEM TO BE SOLVED: To efficiently obtain a compound having an effect for repelling blood- sucking insect pests by treating a substrate with a fungus, etc., having an ability to produce carane-3,4-diol from 3-carene or 3-carane epoxide, and subsequently recovering the product. SOLUTION: This method for producing carane-3,4-diol comprises treating R(+)-3-carene or stereoisomeric 3-carane epoxide having the epoxy ring and the cyclopropane ring at the mutually counter positions with the culture solution of a fungus (e.g. Phaenerochaete chrysosporium IFO-031249) having an ability to produce carane-3,4-diol from the 3-carene or the 3-carane-3,4-diol, the cell collected from the culture solution, or the treated product of the cells, and subsequently recovering the produced (1S, 6R) carane-3,4-diol. The carane-3,4-diol having excellent repelling effects against blood-sucking insect pests such as mosquitoes, gnats, SASHIBAE(a kind of fly), ANDOFURAI(a kind of fly) or NUKAKA(a kind of mosquito), sanitary insect pests such as houseflies, etc. can thus be obtained under the conditions of the ordinary temperature and the atmospheric pressure without requiring a complicated operation.

CONSTITUTION: A cultured solution, fungus body or treated material of the fungus body of a thermophilic microorganism [e.g.; Bacillus smithii SC-J05-1 strain (FERM-BP-4935)] having improved hydration activity converting a nitryl compound to an amide compound, obtained by culturing the microorganism in a medium containing both of a cobalt ion and a manganese ion and having an existing molar ratio of the cobalt ion to the manganese ion of 0.45-2.3 is affected to a nitryl compound (e.g.; acrylonitrile), then the nitryl compound is converted to an amide compound, thus an object amide compound having higher purity than usual (e.g.; acrylamide) is efficiently produced.

Abstract: PURPOSE: To provide a method for producing an amide compound by which the stabler amide compound can be produced even in accumulating the amide compound in a reactional solution at a high concentration. CONSTITUTION: This method for producing an amide compound comprises a step for converting an immobilized microorganism prepared by immobilizing a microorganism having hydrating activities in converting a nitrile compound into the amide compound into particles having 250-1000μm diameter before reacting the nitrile compound with the immobilized microorganism and treating the resultant fine particles with a water-soluble dialdehyde at 0.2-1.0wt.% final concentration in the method for reacting the nitrile compound with the immobilized microorganism and converting the nitrile compound into the amide compound. Furthermore, this method for improving the stability of the hydrating activities in converting the nitrile compound into the amide compound is provided. The immobilized microorganism is improved in the stability of the hydrating activities in converting the nitrile compound into the amide compound.

CONSTITUTION:A cultured solution of a thermophilic microorganism [e.g. Bacillus smithii -SC-J-05-1 (FERM P-14037)] having a hydration activity capable of converting a nitrile compound into an amide compound, its cell body or its treated cell body is added to a nitrile compound (e.g. acrylonitrile) and allowed to act thereon at a temperature range of >= the ordinary temperature to convert this nitrile compound to an amide compound. Since this method can stably convert a nitrile compound into an amide compound even in a temperature range of >= the ordinary temperature, a high-purity amide compound (e.g. acrylamide) can be efficiently synthesized without conducting a low- temperature reaction.
