Abstract:
PURPOSE: A method for preparing arabinose from hemicellulose arabinan is provided to produce functional monosaccharides. CONSTITUTION: A method for preparing L-arabinose comprises a step of immobilizing endo-1,5-alpha-L-arabinanase and alpha-L-arabinofuranosidase to a carrier. A carrier for immobilization is duolite A568. The alpha-L-arabinofuranosidase has an amino acid sequence of sequence number 1. The endo-1,5-alpha-L-arabinanase has an amino acid sequence of sequence number 2. A complex is prepared by immobilizing the enzyme to the carrier. A composition for producing L-arabinose contains the complex.

PURPOSE: A novel mannose-6-phosphoric acid isomerase mutant is provided to obtain L-ribose of high yield and to manufacture pharmaceutical product. CONSTITUTION: A mannose 6-phosphoric acid isomerase mutant includes 90th residue of mannose 6-phosphoric isomerase of sequence number 1, which is substituted with alanine(A), aspartate(D), histidine(H), or leucine(L) instead of aspargine(N); 129th residue which is substituted with phenylalanine(F) or tyrosine(Y) instead of leucine(L); 90th residue which is substituted with alanie(A), aspartate(D), histidine(H) or leucine(L) instead of asparagine(N); and 129th residue which is substituted with phenyl alanine(F) or tyrosine(Y) instead of leucine(L). A composition for preparing ribose comprises the mutant.

PURPOSE: A novel mannose-6-phosphoric isomerase mutant and a method for producing L-ribose are provided to obtain L-ribose of high yield from Geobacillus thermodenitrificans. CONSTITUTION: A novel mannose-6-phosphoric isomerase mutant is prepared by converting 124th amino acid, lysine(K) and 129th amino acid, leucine(L) into arginine(R) and phenyalanine(F). A gene encoding the mutant contains a base sequence of sequence number 4. A method for preparing the mutant isomerase comprises: a step of preparing a recombinant expression vector containing the gene; a culturing a microbe transformed by the expression vector; and a step of isolating the isomerase from the microbe.

PURPOSE: A method for preparing L-ribose using a novel mannose-6-phosphoric isomerase mutant is provided to obtain a large amount of L-ribose from Geobacillus thermodenitrificans. CONSTITUTION: A novel mannose-6-phosphoric acid isomerase mutant is prepared by converting the isomerase 67th amino acid, glutamic acid(E) and 238th amino acid, threonine into glycine(G) and isoleucine(I), respectively. A gene encoding the mutant has a base sequence of sequence number 3. A method for preparing the mutant comprises: a step of preparing a recombinant expression vector containing the isomerase gene; a step of culturing a microbe transformed by the expression vector; and a step of isolating the isomerase from the microbe. 본 발명은 신규한 만노스-6-인산 이성화효소(mannose 6-phosphate isomerase) 효소 및 그 변이 효소 및 그 효소를 이용한 엘-리보스를 제조하는 방법에 관한 것으로, 더욱 상세하게는 만노스-6-인산 이성화효소 및 그 변이 효소 그리고 그 해당 유전자를 포함하는 재조합 발현 벡터, 이로 형질전환된 미생물 및 이들을 이용하여 만노스-6-인산 이성화효소 또는 그 돌연변이체를 대량으로 얻는 제조방법과 상기 만노스-6-인산 이성화효소 또는 그 돌연변이체를 이용하여 엘-리보스를 고수율로 얻는 생산방법에 관한 것이다.
