Abstract: Disclosed herein is a method for synthesizing a capsule by layer-by-layer assembly using dead-end filtration with high operating slurry concentration and high filtered cake concentration, comprising steps of i) providing a core particle, ii) contacting the core particle with a first coating material to form a coated particle slurry having a particle content of at least 4 wt%, ill) filtering the coated particle slurry over an exchangeable filter membrane by dead-end filtration to form a single-layered coated particle cake having a particle content of at least 30 wt%, iv) contacting the coated particle cake with another coating material to form a bi-layered coated particle slurry having a particle content of at least 4 wt%, v) filtering the coated particle slurry over an exchangeable filter membrane by dead-end filtration to form a bi-layered coated particle cake having a particle content of at least 30 wt%, and wherein the coated particle cake is washed by vi) contacting a washing liquid with the coated particle cake to form a washing slurry and vii) filtering it to form a washed coated particle cake. In specific embodiments, the core particle comprises calcium carbonate and an active ingredient i.e. bovine lactoferrin (Lf), coated with protein-polyphenol bilayers, i.e. tannic acid-pepsin bilayers.
