PROBLEM TO BE SOLVED: To provide anti-connexin polynucleotides as impaired wound healing compositions.SOLUTION: There are provided connexin modulation for the treatment of wounds that do not heal at expected rates, including delayed healing wounds, incompletely healing wounds, and chronic wounds, and associated methods, compositions and articles. The present invention provides for increases and improvements in the healing of wounds that do not heal at expected rates, such as delayed-healing wounds, incompletely healing wounds, and chronic wounds, through the use of one or more connexin antisense polynucleotides. In one aspect, the invention comprises a synthetic or naturally occurring wound healing matrix comprising an anti-connexin polynucleotide. In certain embodiments, the anti-connexin polynucleotide is an anti-connexin 43 polynucleotide, for example, an anti-connexin 43 polynucleotide.

PROBLEM TO BE SOLVED: To provide a damaged wound healing composition and therapy.SOLUTION: A method and composition comprising a combination and use of a first anti-connexin agent and a second anti-connexin agent, for example, one or more anti-connexin polynucleotide and one or more anti-connexin peptide or anti-connexin peptidomimetic agent are provided for therapeutic use comprising use for facilitation and/or improvement in wound and wound healing and/or tissue repairing. The present invention encompasses a pharmaceutical composition comprising (a) an anti-connexin peptide or an anti-connexin peptidomimetic agent and (b) a polynucleotide of an antisense to mRNA of connexin protein.

The invention generally relates to methods and compositions for the treatment of a subject suffering from, predisposed to, or at risk for various orthopedic-related diseases, disorders, or conditions. In one aspect, the invention relates to methods of treating of a subject suffering from, predisposed to, or at risk for various orthopedic-related diseases, disorders, or conditions characterized, in whole or in part, by pain, inflammation, excess scarring and/or abnormal tissue formation inside and/or around a joint, comprising a step of administering a composition comprising an anti-connexin agent and a pharmaceutically acceptable carrier or diluent.

The invention generally relates to the use of one or more anti-connexin agents (for example, connexin inhibitors such as α-1 connexin oligodeoxynucleotides and α-1 anti-connexin peptides or peptidomimetics) in combination with one or more therapeutic agents, agents useful for wound healing, and/or gap junction modifying agents for the treatment of wounds, including acute, subacute, delayed healing and chronic wounds.
