PROBLEM TO BE SOLVED: To provide a method for determining information on the three-dimensional structure of a first molecule which forms a complex with a second molecule, with at least one of the molecules being a protein. SOLUTION: The first molecule is made into a complex with the second molecule. At least one of the group of molecules is a protein. The method for determining information on the three-dimensional structure of the first molecule comprises: (a) producing the first molecule in a form substantially labeled with a stable NMR active isotope; (b) forming a complex between the first and second molecules; and (c) subjecting the complex to NMR spectrometry to obtain information on the three-dimensional structure of the first molecule. PROBLEM TO BE SOLVED: To provide a method for preparing isotope-labeled gultamine, asparagine or cysteine used in producing an isotope-labeled protein for NMR measurement, and to provide a prepared product. SOLUTION: This method for preparing the isotope-labeled gultamine or asparagine comprises (a) reacting isotope-labeled gultamic acid or aspartic acid with an N-protecting group to form an N-protected amino acid, (b) cyclizing the N-protected amino acid to form an oxazolidinone, (c) reacting the oxazolidinone with ammonia under an amide formation condition to form a cyclic amide derivative, (d) hydrolyzing the cyclic amide derivative to form an N-protected amino acid, and (e) removing the protecting group from the N-protected amino acid to form the isotope-labeled gultamine or asparagine. PROBLEM TO BE SOLVED: To provide an isotope-labeled nutrient medium for producing an isotope-labeled protein useful for NMR measurement and a method for obtaining a sample for NMR spectroscopy to determine three-dimensional structure information on a protein. SOLUTION: The nutrient medium capable of supporting the proliferation of a mammal or insect cell culture contains all amino acids essential for proliferation of the cells, assimilatable carbohydrate sources, essential inorganic substances, and growth factors.
