    Abstract: Covalently cross-linked pilus polymers displayed on the cell surface of Gram-positive bacteria are assembled by class C sortase enzymes. These pilus-specific transpeptidases located on the bacterial membrane catalyze a two-step protein ligation reaction—first, cleaving the LPXTG motif of one pilin protomer to form an acyl-enzyme intermediate, and second, joining the terminal threonine to the nucleophilic lysine residue residing within the pilin motif of another pilin protomer. Informed by the high-resolution crystal structures of corynebacterial pilus-specific sortase (SrtA) and by developing structural variants of the sortase enzyme whose catalytic pocket has been unmasked by activating mutations, we have developed new reagents capable of forming isopeptide bonds in vitro. The reagents disclosed herein can catalyze ligation of isolated SpaA domains in vitro provide a facile and versatile new platform for protein engineering and bio-conjugation that has major implications for biotechnology.

    Abstract: The present disclosure includes systems and methods for color transfer. The method includes receiving a target image, and determining dominant source colors. The method further includes transforming the target image into a color model including a target luminance component and a target color information component. Additionally, the method includes segmenting the target image into a plurality of target segments based on the target color information component or the target luminance component and extracting dominant target colors from the target image by extracting information for at least one of the dominant target colors from each target segment of the plurality of target segments. Further, the method includes generating a color mapping relationship between the dominant target colors and the dominant source colors, and creating a recolored target image using the color mapping relationship.

    Abstract: The techniques described herein may provide techniques for precise and fully-automatic on-site software failure diagnosis that overcomes issues of existing systems and general challenges of in-production software failure diagnosis. Embodiments of the present systems and methods may provide a tool capable of automatically pinpointing a fault propagation chain of program failures, with explicit symptoms. The combination of binary analysis, in-situ/identical replay, and debugging registers may be used together to simulate the debugging procedures of a programmer automatically. Overhead, privacy, transparency, convenience, and completeness challenges of in-production failure analysis are improved, making it suitable for deployment uses.
