    Abstract: An optical detection assembly for monitoring a biological fluid in a vessel includes two fluid-adjustment structures, which are spaced apart and configured to receive at least a portion of a biological fluid-containing vessel therebetween. A light source (which may be associated with one of the fluid-adjustment structures) is configured to emit light through a thickness of the biological fluid in the vessel, while a light detector (which may be associated with the other one of the fluid-adjustment structures) is configured to receive at least a portion of the light from the light source after it has passed through the biological fluid in the vessel. At least a portion of at least one of the fluid-adjustment structures is configured to move with respect to at least a portion of the other one so as to change the thickness of the biological fluid in the monitored portion of the vessel.

    Abstract: A fluid processing system includes a separation device, a mononuclear cell product processing device, a pump assembly, and a controller. The controller is programmed to actuate the pump assembly to convey blood from a blood source into the separation device, actuate the separation device to separate a mononuclear cell product from the blood, and actuate the pump assembly to convey at least a portion of the mononuclear cell product through the mononuclear cell product processing device to modify a genome of at least one of the cells of the mononuclear cell product and create a modified mononuclear cell product. The controller is programmed to selectively actuate the pump system to convey the mononuclear cell product through the mononuclear cell product processing device in either a first direction or in an opposite, second direction to modify the genome of at least one of the cells of the mononuclear cell product.

    Abstract: A prismatic reflector is provided for incorporation into a centrifugal separation chamber. The prismatic reflector is formed of a light-transmissive material and includes inner and outer walls and first and second end walls. The inner wall is configured to receive light traveling along an initial path and transmit the light to the first end wall, with the first end wall receiving the light transmitted through the inner wall and directing the light toward the second end wall in a direction that is angled with respect to the initial path. The second end wall receives the light from the first end wall and transmits the light out of the prismatic reflector. The initial path of the light may be in a direction toward a rotational axis of the centrifugal separation chamber, with the prismatic reflector redirecting the light into a direction substantially parallel to the rotational axis.

    Abstract: An umbilicus-driven centrifuge includes a yoke configured to orbit a midsection of the umbilicus around a rotational axis at a first speed so as to cause a separation chamber associated with the umbilicus to rotate about the rotational axis at a second speed that is approximately double the first speed. An optical monitoring system directly monitors the separation chamber, with a light source oriented to emit light toward the separation chamber and a light detector oriented to receive at least a portion of the light after the light has passed through the separation chamber. A controller receives a plurality of signals from the light detector, then compares the period or frequency of the signals to an expected period or frequency. The controller determines that the umbilicus is experiencing an irregularity when the period or frequency is different from the expected period or frequency.

    Abstract: Systems and methods are provided for determining and controlling the location of an interface between separated blood components within a centrifuge. An optical monitoring assembly is used to determine the position of the interface, with the current position being compared to a target position. An error signal, which is indicative of the distance between the two positions, is input into a proportional-integral-derivative control algorithm to generate a control signal. The control signal is used to modify the rate at which a separated blood component is removed from the centrifuge so as to move the interface toward the target position. Blood characteristics, blood source characteristics, and/or the rate at which blood is pumped into the centrifuge are monitored for any changes, with a change in one or more of these factors being used to modify the control algorithm.

    Abstract: Biological fluid processing cassettes with integrated filter structures are disclosed. A cassette body is formed of a generally rigid material, defining a plurality of internal fluid flow paths. The cassette body may be secured to a cassette cap to define a cavity, with a filter sealed within the cavity. Each of the cassette body and the cassette cap defines a port opening into the cavity, which allows fluid to flow from one of the internal fluid flow paths, into the cavity and through the filter, and then out of the cavity via the port of the cassette cap. Alternatively, the cassette body may define an external slot which receives at least a portion of a filter. Such a filter includes two ports, with one of the ports in fluid communication with a cassette port of the cassette body to allow fluid flow between the cassette body and the filter.
