Abstract: Embodiments of the invention relate generally to protein extraction and, more generally, to bone protein extraction methods that do not require demineralization. In one embodiment, the invention provides a method comprising: mixing a bone sample and a quantity of an extraction buffer comprising: ammonium phosphate dibasic; or ammonium phosphate dibasic and ammonium bicarbonate; or ammonium phosphate dibasic, ammonium bicarbonate, and guanidine HCl; or sodium phosphate dibasic and sodium bicarbonate; or sodium phosphate dibasic, sodium bicarbonate, and guanidine HCl; or potassium phosphate dibasic and potassium bicarbonate; or potassium phosphate dibasic, potassium bicarbonate, and guanidine HCl; and incubating the bone sample/extraction buffer mixture.

Abstract: A method of predicting bone fracture risk in a type 2 diabetes (“T2D”) patient includes: obtaining a plurality of averaged glycated hemoglobin (“HbA1c”) values for a plurality of T2D subjects over an observational period; binning the plurality of averaged HbA1c values into two predetermined categories; obtaining a plurality of total fracture incidences for the plurality of T2D subjects over a follow-up period; performing a plurality of linear regression model correlations between the binned HbA1c values and the plurality of total fracture incidences to determine a bone fracture rate prediction model; obtaining a measurement of an HbA1c value of the T2D patient; and analyzing the HbA1c value of the T2D patient with the bone fracture rate prediction model to determine the T2D patient's risk of bone fracture for a future period.

Abstract: Provided is a method of mineralizing tissue, including placing within a body of a subject a substance, wherein the substance includes osteopontin and osteocalcin. Also provided is a tissue mineralization-promoting substance including osteopontin and osteocalcin, and an article for promoting tissue mineralization, including an implant which may be an orthopedic implant or an endosseous implant, and a substance disposed on a surface of the implant including osteopontin, and osteocalcin. A method of making a tissue mineralization-promoting substance, including combining osteopontin and osteocalcin, is also provided. Other features, including calcium phosphate, type-I collagen, mesenchymal stem cells, and growth factors, are also provided.
