Abstract:
A surface-enhanced Raman scattering nano-tagging particle and a preparation method thereof are provided to reduce the preparation costs of the particle by using silica, and to detect biological molecules economically and rapidly. The surface-enhanced Raman scattering nano-tagging particle comprises a silica core particle having a nano silver particle immobilized with a tagging material and a silica skin precursor in the surface, and a silica skin enclosing the silica core particle, wherein the size of the silica core particle is 50-300nm. The tagging material is 2-methylbenzenethiol, 4-methylbenzenethiol, 4-mercaptopyridine, 2-naphthalenethiol, 4-methoxybenzenethiol, 3-methoxybenzenethiol, 3,4-dimethylbenzenethiol, thiophenol or 3,5-dimethylbenzenethiol, and the silica skin precursor is 3-mercaptopropyltrimethoxysilane or 3-aminopropyltriethoxysilane. The silica skin is formed by tetraethyl orthsilicate and sodium silicate and has the thickness of 10-50 nm. The method for preparing the surface-enhanced Raman scattering nano-tagging particle comprises the steps of: introducing nano silver particle on the surface of silica core particle; immobilizing the tagging material and silica skin precursor to the nano silver particle; and forming the silica skin around the silica core particle immobilized with the tagging material and silica skin precursor.

본 발명은 생체분자 고속 검출에 사용될 수 있는 표면 증강 라만 산란 나노-표지 입자 및 이의 제조방법에 관한 것이다. 본 발명에 따른 표면증강 라만 산란 나노-표지 입자는 매우 간편하게 제조될 수 있으며, 라만 스펙트럼에 매우 민감하기 때문에 분석이 용이하다는 이점이 있다. 또한, 표면에 표지될 수 있는 화학물질로 다양한 종류가 사용될 수 있기 때문에, 매우 경제적으로 제조될 수 있다. Abstract:
A method for selecting and analyzing compounds or biomolecules is provided to more economically screen many compounds and biomolecules with high speed by using microbead labeled by surface-enhanced Raman scattering and dielectrophoresis. The method comprises the steps of: (a) labelling micro-beads with nano-silver particles and a chemical material showing high binding force with the nano-silver particles such as 2-methylbenzene thiol, 4-methylbenzene thiol, 2-naphthalene thiol, 4-methoxybenzene thiol, 3-methoxybenzene thiol, 3,4-dimethylbenzene thiol, 3,5-dimethylbenzene thiol, 2-mercapto toluene, 4-mercapto toluene and 4-mercapto pyridine; (b) introducing a biomolecule-specific ligand such as biotin, antibody, lectin and peptide into the labelled micro-bead surface; (c) introducing a biomolecule into the ligand-introduced microbead; (d) selecting the biomolecule-introduced microbead using dielectrophoresis; and (e) analyzing the selected biomolecule through surface-enhanced raman spectroscopy.
