Abstract:
본 발명은 세포투과성 펩타이드인 Pep-1을 이용한 신규한 항균 펩타이드 Pep-1-K 및 그 용도에 관한 것으로, 구체적으로 세포투과성 펩타이드로 알려진 Pep-1의 2번째, 6번째 및 11번째의 아미노산인 글루탐산(Glu)을 라이신(Lys)으로 치환시켜 제조한 항균 펩타이드 Pep-1-K 및 그 용도에 관한 것이다. 본 발명의 Pep-1-K는 다양한 균주, 그 중에서도 그람-양성균, 그람-음성균 및 항생제에 내성을 가진 균주에서 강력한 항균활성을 나타낼 뿐 아니라 인간 적혈구 세포에서 어떠한 용혈활성도 나타내지 않기 때문에 무독성 항균제, 식품 방부제 및 화장품 첨가제로 유용하게 사용될 수 있다. 세포투과성 펩타이드, Pep-1, Pep-1-K
Abstract:
A novel synthetic or a recombinant antibacterial peptide is provided which shows strong antibacterial activity against various strains such as gram positive bacillus, gram negative bacillus and a strain having the resistance against antibiotics without any hemolytic activity in human erythrocyte cells, thereby being usefully used as a non-toxic antibacterial agent, a food preservative and a cosmetic additive. A synthetic or a recombinant antibacterial peptide is characterized in that it prepared by substituting at least one Glu at the 2nd, the 6th and the 11th of a cell-penetrating peptide Pep-1 with a positively charged basic amino acid such as arginine(Arg), histidine(His) and lysine(Lys) and it has the antibacterial activity against at least one strain selected from the group consisting of Bacillus subtilis KCTC 3068, Staphylococcus epidermidis KCTC 1917 and Staphylococcus aureus KCTC 1621. A non-toxic antibacterial agent comprises the antibacterial peptide as an effective ingredient.
Abstract:
A model type antibiotic peptide to have abundant tryptophan is provided to show very low hemolytic activity in human RBC, to show very high bacterial cell specificity, to show high stability to human serum and to be usefully used as halogen free antimicrobial agent, food antiseptic and make-up additive. A synthesis or recombining antimicrobial peptide is selected from a group consisting of i) peptide selected from a group consisting of sequence number 1 to 4, ii) peptide in which lysine(Lys) is substituted for ornithine(Orn) from the peptide of i), iii) peptide in which leucine(Leu) is substituted for deer leucine(Nle) from the peptide of i) and iv) enantiomer peptide substituted for D-type amino acid corresponding all amino acids from the peptide of i) to iii). The peptide of ii) is written as sequence number 5.
