Abstract: A method for preparing hair follicle stem cells is provided to use the obtained stem cells capable of being differentiated into hair follicle cells and growing hair as a new cell therapeutic agent of hair loss. A method for isolating hair follicle stem cells and differentiating them into hair follicle cells comprises the steps of: (a) after finely incising scalp tissue including hair follicle, performing a first chemical decomposition of the tissue in a culture medium comprising a protease, a collagenolytic enzyme and a DNAse enzyme; (b) performing a second chemical decomposition on the tissue obtained from the step(a) in a culture medium comprising collagenase; (c) after recovering the chemically decomposed tissue and culturing scalp cells in a culture medium including 1-30% of serum, culturing it in a non-serum culture medium after cell attachment; (d) after recovering the cultured scalp cells, isolating CD34+ therefrom and isolating hair follicle stem cells; and (e) hypodermically injecting the isolated hair follicle stem cells into the scalp tissue to be differentiated into hair follicle cells, wherein the culture medium for the first chemical decomposition comprises 1-10% of an enzyme complex consisting of 0.1-2mg/ml of dispase, protease, collagenolytic enzyme and DNAse enzyme in a DMEM culture medium including 1-30% of fetal bovine serum, 10-200mug/ml of normocin, and 1-10% of penicillin-streptomycin and the culture medium for the second chemical decomposition comprises 0.1-10mg/ml of collagenase type I A in the DMEM culture medium. A composition for promoting hair growth comprises the hair follicle stem cells as an effective ingredient.
