    Abstract: A recombinant microorganism is provided to secret a highly concentrated human epidermal growth factor in a pure state into a cell culture solution, thereby isolating and purifying the human epidermal growth factor very efficiently. A recombinant microorganism is co-transformed by a recombinant vector including a gene encoding an outer membrane protein F(OmpF) derived from E. coli and a recombinant vector including a gene encoding a human epidermal growth factor and secreting and expressing the human epidermal growth factor into a periplast of microorganism, thereby the gene encoding the OmpF and the gene encoding the human epidermal growth factor being expressed simultaneously and the human epidermal growth factor being extracellularly produced. A method for extracellular production of the human epidermal growth factor comprises the steps of: (a) culturing the recombinant microorganism to secret the human epidermal growth factor into a culture solution; and (b) collecting the human epidermal growth factor from the culture solution.

    Abstract: PURPOSE: A method for enhancing expression of a target protein through co-expression of tRNA of a specific amino acid is provided to enhance productivity of specific amino acid. CONSTITUTION: A method for enhancing expression of a target protein is co-expression of a gene encoding the target protein and gene sequence encoding tRNA of a specific amino acid using recombinant microorganism. The recombinant microorganism is obtained by transforming with the gene encoding the target protein and gene sequence encoding tRNA. The target protein is repetitive protein having repetitive structure of a specific oligopeptide. The repeat protein is elastin, silk protein, byssus, flagelliform silk, dragline silk, collagen, keratin, sericin or synthetic repetitive protein. The recombinant microorganism is E.coli. The recombinant vector contains T7 promoter.

    Abstract: A new promoter form expressing and secreting a target protein is provided to produce pure target protein by extracellularly expressing the target protein. Gene constructs comprise: a secretion promoting motif gene selected among a group of a coding gene of arginine transporter subunit(ArtJ), a coding gene of hyperosmotically inducible periplasmic protein(OsmY) and a coding gene of peptidyl-prolyl cis-trans isomerase(FkpA) and a gene coding a target protein. The gene constructs are characterized in intracelluarly expressing in a fusion form of the secretion promoting motif selected and the target protein and extracelluarly secreting. Method of extracellular secretion and production of the target protein comprises: a step of culturing recombinant microorganism containing the gene constructs to extracellularly secret the target protein; and a step of collecting the target protein in culture media.
