The invention relates to a microplate reader and a respective method, wherein the microplate reader comprises at least one measuring device and a holding device for accommodating at least one microplate and for positioning the samples-containing wells of this(these) microplate(s) in relation to the at least one measuring device. The at least one measuring device is used for detecting light which is emitted by samples in wells of a microplate inserted in this microplate reader and/or which is influenced by samples transilluminated by light in wells of a microplate inserted in this microplate reader. The microplate reader comprises a control unit for controlling the temperature of a gas atmosphere surrounding the wells containing the samples of microplates used in this microplate reader.

A system for detecting emissions from a sample containing nucleotide molecules contains a sample holder, an optical illumination system, an optical sensor. The sample holder includes a plurality of spatially separated reaction sites configured to hold a sample containing nucleotide molecules. The optical illumination system comprising a radiant source configured to simultaneously illuminate two or more of the reaction sites. The illumination system includes a homogenizer. An output from the homogenizer has less variation in power, energy, irradiance, or intensity than the variation in power, energy, irradiance, or intensity of the source.

A fluorescence detection apparatus for analyzing samples located in a plurality of wells in a thermal cycler and methods of use are provided. In one embodiment, the apparatus includes a support structure attachable to the thermal cycler and a detection module movably mountable on the support structure. The detection module includes one or more channels, each having an excitation light generator and an emission light detector both disposed within the detection module. When the support structure is attached to the thermal cycler and the detection module is mounted on the support structure, the detection module is movable so as to be positioned in optical communication with different ones of the plurality of wells. The detection module is removable from the support structure to allow easy replacement.

A unit is provided comprising an array (2) of sample containers (1), said containers, being connected together and arranged in a planar configuration, each container having multiple optically transparent windows arranged such that the sample contained therein can be interrogated using simultaneous multiple optical analytical techniques, the array of containers being configured so as to allow optical access to the windows of each container in the array. Also provided is an apparatus comprising such a unit, a system comprising a combination of such an apparatus and unit and a method of analysing multiple samples by introducing each individual sample into an individual container of such an apparatus, illuminating the samples and detecting and analysing light emerging therefrom.

A system and method for fluorescence excitation and detection having distinct optical paths is disclosed. A system for detecting fluorescence comprises a light source that emits an excitation light into an illumination tube; a plurality of collection optics located around an aperture in the illumination tube for collecting fluorescence; and a detector for determining the amount of fluorescence. A method for detecting fluorescence comprises emitting an excitation light from a light source into an illumination tube; directing the excitation light to an excitation filter; illuminating a sample with the excitation light to generate an emission light; and detecting the optical characteristics of the emission light using a plurality of collection optics located around the illumination tube.

The invention relates to a method of performing a fluorescence assay. According to the invention a filter is integrated with at least one component chosen from i) a body comprising a well, wherein the well in which the assay is being performed is exposed to excitation light in such a manner that the filter is positioned between the light-sensitive element and the excitation light source, and ii) a light-sensitive element, wherein the filter is applied at least at the light-sensitive side of the surface of the light-sensitive element, and any light that may be emitted is detected by means of the light-sensitive element. The invention also relates to an apparatus suitable for carrying out the method, as well as a method for its manufacture.
