Abstract:
PURPOSE: A two-photon dye is provided to ensure high selectivity for Zn^2+ and to enable monitoring effectively of intracellular free zinc ions for a long time, wherein Zn^2+ within cytoplasm is positioned deeply from the surface. CONSTITUTION: A two-photon dye for real time monitoring of intracellular free zinc ions has a structure of formula 1. A method for preparing the two-photon dye comprises (a) mixing a compound of chemical formula 2 into an organic solvent, and stirring the materials to obtain a first mixture; and (b) synthesizing a compound of chemical formula 4 by adding a compound of chemical formula 3 to the first mixture, and then reacting the materials.
Abstract:
A two proton probe is provided to show excellent ion selectivity on magnesium and very high two proton fluorescence efficiency, thereby being adequate for real time image of intracellular magnesium ions. A two proton probe for real time monitoring of intracellular magnesium ions is represented by a formula(1) and is prepared by reacting a compound represented by a formula(2) with 1-ethyl-3-(3-dimethylaminopropyl)-carbodiimide.HCl, a compound represented by a formula(3) and 4-dimethyl aminopyridine. In the formula(1), R is H or CH2OCOCH3. A method for real time monitoring of intracellular magnesium ions comprises a step of injecting the two proton probe into cytoplasm to be subject to observation and then observing a two probe excited fluorescence image emitted therefrom. Further, the two probe excited fluorescence image is obtained by a wavelength ranged from 500 to 620nm.

본 발명은 유의미한 이광자 작용 단면 및 pH에 반응하여 청색-대-녹색 발광 색상의 뚜렷한 변화, 높은 광안정성과 낮은 세포독성을 보이므로 생체내의 pH를 효과적으로 측정할 수 있는 이광자 표지자를 제공한다. 본 발명에 따른 이광자 표지자는 이광자 현미경법을 통한 비율계량법에 의해 인간의 위와 식도 조직 뿐 아니라 생세포에서 90-180㎛ 깊이의 pH를 효과적으로 측정할 수 있다.
