Abstract:
A method for isolating magnesium lithospermate B from Salviae miltiorrhizae is provided to improve isolation yield and purification efficiency of magnesium lithospermate B with use of expensive isolation materials such as sephadex LH-20, MCI gel CHP 20 P and silica gel, so that magnesium lithospermate B is useful to treat and prevent diabetic nephropathy, microalbuminuria and uremia. A method for isolating magnesium lithospermate B from Salviae miltiorrhizae comprises the steps of: (a) cutting the roots of Salviae miltiorrhizae and extracting them with methanol at room temperature for 3 days; (b) suspending the crude extract of Salviae miltiorrhizae with water, filtering and concentrating the water extract of Salviae miltiorrhizae at 70 deg. C, and regulating pH of the water extract with 10% HCl to 3.3-3.7; (c) adding hexane into the concentrated water extract to remove organic matters, and washing the purified water extract with ethylacetate to divide into ethylacetate extract and water extract; (d) regulating pH of the water extract with 10% HCl to 4.3-4.7, adding butanol into the water extract, removing solvent from the water extract at 70 deg. C, and drying the water extract under vacuum for 5 hours to obtain pure magnesium lithospermate B; and (e) adding distilled water into the ethylacetate extract, regulating pH of the ethylacetate extract with 10% HCl to 4.3-4.7, extracting the ethylacetate extract with new ethylacetate, and concentrating and drying the ethylacetate extract to obtain magnesium lithospermate B.
