Abstract:
본 발명은 부탄올 생성능이 증가된 재조합 미생물 및 이를 이용한 부탄올의 제조방법에 관한 것으로서, 더욱 상세하게는 대사회로의 조작을 통하여 부탄올 생성능이 증가된 재조합 미생물 및 이를 이용한 부탄올의 제조방법에 관한 것이다. 본 발명에 따른 부탄올 생성능이 증가된 재조합 미생물은 아세틸코에이(acetyl CoA) 및 부티릴코에이(butyryl CoA)로의 생합성 경로에 관여하는 효소들을 코딩하는 유전자를 가지는 숙주 미생물에서, 아세틸코에이(acetyl CoA)를 아세테이트로 전환하는 효소를 코딩하는 유전자가 결실되어 있는 것을 특징으로 한다. 미생물의 대사흐름 조작에 의해 수득된 본 발명에 따른 재조합 미생물은 부탄올을 선택적으로 고효율로 생성할 수 있어, 산업 용제 및 수송 연료 생성 미생물로 유용하다.
Abstract:
PURPOSE: A recombinant microorganism containing a gene encoding an ACS/CODH(acetyl-CoA synthases/carbon monoxide dehydrogenase) complex is provided to ensure carbon dioxide fixation ability, and to produce high concentratioin and high yield useful substances such as alcohol and organic acid. CONSTITUTION: A recombinant microorganism with a carbon dioxide fixation ability contains a gene encoding ACS/CODH complex or a recombinant vector containing the gene in a host microorganism. A method for preparing the recombinant microorganism comprises a step of introducing the gene or the recombinant vector into the host microorganism. The method for fixing carbon dioxide comprises a step of culturing the recombinant microorganism under the presence of CO_2. A method for preparing the useful substance comprises: a step of culturing the recombinant microorganism using CO_2 as a carbon source; and a step of generating and collecting the useful substance. The useful substance is alcohol including butanol or ethanol.

PURPOSE: A method for inactivating Ralstonia sp. microorganism gene is provided to enable genetic study of the microorganism. CONSTITUTION: A recombinant vector contains: sacB gene encoding evansucrase of Bacillus subtilis, a selective marker, a base sequence for selective marker removal; and a sequence for homology recombination. The recombinant vector additionally contains origin of replication. The recombinant vector additionally contains a gene which enables DNA transition by hybridization between cells. The selective marker exists between the sequences causing homology recombination with a target gene.

PURPOSE: A mutant microorganism which prevents productivity reduction of a target material is provided to prevent degeneration of the microorganism regardless of medium and fermentation condition. CONSTITUTION: A mutant microorganism which prevents productivity reduction of a target material by megaplasmid deletion is prepared by a part of inserting essential chromosome DNA and complementary site to a microorganism with the megaplasmid. The part of the chromosome DNA is a gene encoding DNA polymerase, RNA polymerase, or ribosome.
