Abstract: PURPOSE:The titled cells, containing a DNA sequence coding a human tissue plasminogen activation factor (t-PA) produced by human normal cells, capable of establishing a mass-production process without hazard and useful for treating thrombosis. CONSTITUTION:Human normal diploid cells having a high ability to produce human tissue plasminogen activation factor (t-PA), 2n=46 number of chromosome and limited passage number, e.g. MTC-017, are cultivated to extract mRNA from the resultant cultivated cells. A fraction coding t-PA is sorted out of the mRNA and the resultant fraction is converted into double stranded cDNA by a coupler method, etc. The resultant cDNA is then inserted into a plasmid PUC13 and then transformed in Escherichia coli. DNA (A) is then separated from the resultant cells. On the other hand, the t-PA is separated from the MTC-017 strain to synthesize a nucleotide (B) complementary to the DNA. The components (A) and (B) are hybridized to give a hybrid, from which a vector containing the DNA sequence, coding the t-PA and expressed by the formula is obtained. Animal cells are transformed with the vector and the cells thereof are then separated.

The therapeutic agent increases the production of interferon and macrophage-activating factors, and activates NK(natural killer) cells and macrophages. The agent further increases the production of colony- stimulating factors and accelerates the multiplication of lymphocytes.
