Abstract:
A method for providing an adenovirus from a serotype which does not grow efficiently in a desired cell line with the ability to grow in that cell line is described. The method involves replacing the left and right termini of the adenovirus with the corresponding termini from an adenovirus which grow efficiently in the desired cell line. At a minimum, the left terminus spans the (5′) inverted terminal repeat, the left terminus spans the E4 region and the (3′) inverted terminal repeat. The resulting chimeric adenovirus contains the internal regions spanning the genes encoding the penton, hexon and fiber from the serotype which does not grow efficiently in the desired cell. Also provided are vectors constructed from novel simian adenovirus sequences and proteins, host cells containing same, and uses thereof.

Sequences of a serotype 8 adeno-associated virus and vectors and host cells containing these sequences are provided. Also described are methods of using such host cells and vectors in production of rAAV particles. rAAV8-mediated delivery of therapeutic and immunogenic genes, including Factor VIII, is also provided.

Mass storage system controller apparatus receives requests for access to files in a data storage system and manages access to the files in the data storage system, the data storage system having a plurality of storage devices and the requests being addressed to the data storage system as a single virtual storage device having addressable Command Regions allocated to controller commands and addressable Mapped Data Regions allocated for data storage. The apparatus includes a file system to manage access to files in the data storage system, the file system having a processor acting as a Command Region processor and acting as a Mapped Data Region processor. A disk emulator responds as a disk drive device to write/read commands and passes the write/read commands addressed to Command Regions of the virtual storage device to the Command Region processor, the write/read commands addressed to Command Regions containing controller commands for the file system. The Command Region processor responds to controller commands in the write/read commands and processes the controller commands to control files in the data storage system. The disk emulator responds as a disk drive device to write/read commands and passes write/read commands addressed to a Mapped Data Region of the virtual storage device to the Mapped Data Region processor. The Mapped Data Region processor responds to a write/read command with a Mapped Data Region address and transfers data to or from the data storage system for files mapped to the Mapped Data Region address.

The invention provides a recombinant viral vector comprising the DNA of, or corresponding to, at least a portion of the genome of an adenovirus, which portion is capable of infecting a hepatic cell; and a human VLDL receptor gene operatively linked to regulatory sequences directing its expression. The vector is capable of expressing the normal VLDL receptor gene product in hepatic cells in vivo or in vitro. This viral vector is useful in the treatment of metabolic disorders caused by the accumulation of LDL in plasma, such as familial hypercholesterolemia or familial combined hyperlipidemia.

A heating element adjusts the curvature of a cylindrical wobble correction mirror to compensate and correct the scan line bow for an optical scanner. A current applied through the heating element along one side of the mirror will cause the mirror to bend vertically for a horizontal beam to adjust the scan line bow for a single beam and approximately equalize the scan line bow for multiple beams.
Abstract:
A honeycomb structure is formed integral with a raster output scanning system housing. A constrained layer damper is bonded to the honeycomb structure and the raster output scanning system is mounted on the constrained layer damper. The honeycomb structure and constrained layer damper provide support and reduce vibrations to the raster output scanning system. The cells of the honeycomb structure can be irregular in height, thickness, density or shape to further support and damp vibrations.

Methods for efficient production of recombinant AAV employ a host cell which comprising AAV rep and cap genes stably integrated within the cell's chromosomes, wherein the AAV rep and cap genes are each operatively linked to regulatory sequences capable of directing the expression of the rep and cap gene products upon infection of the cell with a helper virus, a helper gene, and a helper gene product. A method for producing recombinant adeno-associated virus (rAAV) involves infecting such a host cell with a helper virus, gene or gene product and infecting the infected host cell with a recombinant hybrid virus or plasmid vector containing adenovirus cis-elements necessary for replication and virion encapsidation, AAV sequences comprising the 5′ and 3′ ITRs of an AAV, and a selected gene operatively linked to regulatory sequences directing its expression, which is flanked by the above-mentioned AAV sequences.

Methods for efficient production of recombinant AAV are described. In one aspect, three vectors are introduced into a host cell. A first vector directs expression of cre recombinase, a second vector contains a promoter, a spacer sequence flanked by loxP sites and rep/cap, and a third vector contains a minigene containing a transgene and regulatory sequences flanked by AAV ITRs. In another aspect, the host cell stably or unducibly expresses cre recombinase and two vectors carrying the other elements of the system are introduced into the host cell.
Abstract:
The present invention provides a hybrid vector construct which comprises a portion of an adenovirus, 5′ and 3′ ITR sequences from an AAV, and a selected transgene. Other hybrid vectors form a polycation conjugate and incorporate an AAV rep gene in a single particle. These hybrid virus vectors are characterized by high titer transgene delivery to a host cell and the ability to stably integrate the transgene into the host cell chromosome. Also disclosed is the use of the hybrid vectors to produce large quantities of recombinant AAV.
