Abstract: PURPOSE: To reduce deactivation of enzyme during preservation, by adding modified gelatin to an enzyme-containing solution, lyophilizing it.
CONSTITUTION: Gelatin is heat-treated at 80W120°C for 5W20min, to give modified gelatin. The modified gelatin has 5,000W1,000 average mol.wt. The modified gelatin is added to an enzyme-containing solution in such a way that powder prepared after lyophilization contains 5W80wt% (especially 10W60wt%) modified gelatin, and it is melted at a temperature not to deactivate the enzyme. It is then lyophilized by a conventional procedure.

Abstract: PURPOSE:To analyze glucose or cholesterol quantitatively with accuracy, without reaction disturbance due to air, etc., by maintaining the temperature of a buffer bath higher than that of the reaction vessel. CONSTITUTION:The buffer 6, maintained at a temperature about 5-10 deg.C higher than that of the reaction vessel 16 by the heater 2 of the buffer bath 1, is sent to the sample injection unit 8 by the transfer pump 7. A given amount of a sample, e.g., serum, is injected by the syringe 10 into the buffer 16, which is then passed from the pipe 11 through three-way cock 14 to the vessel 16 at about 25-40 deg.C in the thermostatic chamber 20, and reacted with the immobilized oxidase set therein. After completion of the reaction, the buffer 16 is passed from the pipe 17 to the oxygen analyzer 18 to determine the consumed oxygen and amount of glucose and cholesterol. The liquid after the determination is passed from the pipe 19 through the three-way cock 15, and discharged from the pipe 13.

Abstract: PROBLEM TO BE SOLVED: To obtain a urea determination reagent for subjecting urea derived from a biological substance to a rating assay by using a urease reaction as a rate-determining step without highly diluting the sample by using a variant type urease having reduced affinity for urea. SOLUTION: A natural urease gene derived from Klebsiella aerogenes is subjected to site-directed mutagenesis so that histidine at the amino acid sequence α 219th of urease is mutated into alanine, asparagine, glutamine, etc., to give a variant type urease having reduced affinity for urea. The variant type urea having reduced affinity for urea is used to give the objective urea determination reagent capable of determing urea derived from a biological substance by using a urease reaction as a rating assay without highly diluting the sample and without adding a substance for regulating the affinity of enzyme to a substrate by directly suppressing urease.

Abstract: PURPOSE:To obtain a diaphragm type gaseous carbonic acid sensor excellent in sensitivity and stability and capable of calibrating by a pH buffer solution in good reproducibility, by using a reticulate nonwoven cloth as a spacer between a gas selectively permeable diaphragm and an electrode membrane. CONSTITUTION:A reticulate nonwoven cloth spacer 2 is attached between a gas selectively permeable diaphragm 1 of a sensor consisting of an outer cylindrical pipe 5 providing with a gas selectively permeable diaphragm 1 at the one end, a composite pH electrode 6, and an inner liquid 7 and a composite pH electrode 6 by a spacer band 4. Synthetic fibers such as polyamide, polyester etc. are used for materials of the reticulate nonwoven cloth spacer but hydrophillic materials such as cotton, rayon etc. are preferable from a viewpoint of being accustomed to an electrolyte. The reticulate nonwoven fabric has a good permeability to the inner liquid and gaseous carbonic acid is diffused easily and also, an output is stable. Further, sensitivity is good and meshes are not disturbed by detaching and attaching the space and also, the reproducibility of measured value is good.

Abstract: PURPOSE:To raise safety by installing a device that detects the flow of a buffer solution and cutting off the power source to temperature regulating device and so on or sounding an alarm buzzer or other safety means. CONSTITUTION:A buffer solution 16 in a vessel 15 for the buffer solution is supplied by means of a circulation pump 25 through a pipe 17 to a pipe 27 that is wound round a heating drum 26. The buffer solution whose temperature has reached a constant temperature through the heating drum 26 is returned again to the buffer solution vessel 15 by traversing the wall. Here the oxygen dissolved in the buffer solution is balanced by the multiple effect of the negative effect to the buffer solution and pulsating flow effect by the circulation pump 25 and the heating effect by the heating drum 26. While the buffer solution is circulating, the light flux from the light source 19 gets into a light receiving element 21 to turn on a switching circuit and supplies electric power of the power source 24. If the buffer solution is exhausted, total reflection develops by the difference of the reflective indices by passing from a glass tube 20' to the air and incident light to the light receiving element 21 to turn off the switching circuit and overheat is avoided.
