Two-photon dye for real-time monitoring of cells, in which the charge transport within molecule is facilitated because of having high deltaTPA by holding strong donor-acceptor pairs within the annular planar structure, the optical stability is excellent because of having the conjugated bond within the loop, the water solubility is excellent because of having the integrated ester as receptor, and the cell permeability is superior because of having a small molecular weight, is provided. And a preparing method of the two-photon dye and a real-time cell monitoring method using the same are also provided. Two-photon dye for real time monitoring of cells is represented by a formula 1. In the formula 1, X is -OCH3, and -OH or - N(CH3) 2, and Y is -COOCH3, and -CN or -COOC4H4NO2. The maximum wavelength of the two-photon excitation spectrum of the compound in the formula 1 ranges from 780nm to 940nm.

Abstract:
A two-photon dye for real-time imaging of lipid raft is provided to realize high solubility in water and high fluorescence intensity under an environment inside cellular membranes without causing cytotoxicity, and to enable direct visualization and three-dimensional real-time imaging of lipid raft. A two-photon dye for real-time imaging of lipid raft is a compound represented by the following formula 1. In formula 1, R1 is a C5-C17 alkyl group; R2 is -COOH, -CH2SO3Na or the formula (a); and R3 is a C5-C17 alkyl group. The two-photon dye for real-time imaging of lipid raft has an in vivo two-photon absorptivity of at least 20 GM, and the two-photon excitation occurs under a light source with a wavelength range of 750-1000nm and fluorescence occurs at a wavelength of 400-700 nm.
