A single sample quench determination method including generating a pulse height distribution spectrum for the sample in the presence of a standard source, measuring a pulse height value corresponding to a unique point on the spectrum, counting all pulses produced by a radionuclide within the sample, counting pulses produced by the radionuclide in the sample within a calibration window, optically varying the quench of the single sample and repeating such steps, correlating normalized pulse height values to the counts for all pulses produced by the radionuclide within the sample at varying degrees of quench to determine disintegrations per minute within the single sample, determining efficiency for the pulse counts measured within the calibration window, and relating such efficiencies to an indication of quench to thereby provide a quench relationship from a single sample.
Abstract:
A system for cooling and lubricating the drive motor of a centrifuge. The shaft (16) of the motor (10) includes an internal passage (40) with an inlet (48) having a size and taper sufficient to provide the desired rate of cooling oil flow through the motor. A lower bearing proportioning arrangement (40) applied a high percentage of a small proportion of the total oil flow to the lower bearing (20). An upper bearing proportioning arrangement (50) applies a low percentage of a large proportion of the total oil flow to the upper bearing (18). Together these features assure a high degree of motor cooling without overlubricating the bearings.
Abstract:
An electrophoretic procedure of the type comprising (a) applying at least one sample to a gel; (b) electrophoresing the gel of step (a); (c) fixing the electrophoresed gel of step (b); and (d) drying the fixed gel of step (c). The electrophoretic procedure is characterized in that a novel electrophoretic gel is employed therein. The novel electrophoretic gel is of the type comprising (a) at least one marine hydrocolloid processed from the class Rodophycea; and (b) a buffer. The electrophoretic gel is characterized in that the buffer (i) is selected from a group consisting of adipic acid, glutaric acid, itaconic acid, maleic acid, malic acid, malonic acid, succinic acid, succinamic acid, and tricarbalyllic acid and (ii) is present in a concentration of about 0.05 to about 0.09 M.

An improved mirror scan control for driving a movable mirror (14) in an interferometer with a constant scan velocity comprises a closed loop servo control which provides constant velocity mirror scan in response to a phase comparison of a signal derived from the beat frequency of a two frequency laser beam (16) passing through the interferometer and a selected frequency reference signal, wherein the difference in frequencies or beat frequency of the two frequency laser beam is stabilized at a specified difference in frequencies. The mirror scan control employs a phase lock control loop which locks the frequency of the referenced signal with the frequency of the signal derived from the laser beam to provide precise mirror velocity control.

An improved apparatus for sequencing proteins and peptides in a reaction chamber using a spinning cup by successive coupling and cleavage reaction steps wherein the reaction chamber is always subjected to positive pressure. The improvement involves the elimination of the use of a vacuum in the chamber for drying or enhancing reagent or solvent transfer. Throughout the coupling and cleavage steps the reaction chamber is subjected to a positive pressure by the positive application of an inert gas through the reaction chamber for reagent and solvent transfer, drying and removal of waste.

A method for assaying the direct bilirubin content of a sample comprising (a) mixing a sulfanilic acid solution comprising from about 0.02 to about 0.05 M sulfanilic acid and from about 0.02 to about 0.15 M hydrochloric acid with a sodium nitrite to thereby commence a diazotation process, the ratio of the sulfanilic acid solution to said sodium nitrate solution being preselected so as to yield a diazo solution comprising from about 0.0001 to about 0.001 M Diazo dye; (b) allowing the diazotation process to form the diazo solution for at least 15 seconds; (c) introducing an aliquot of the sample to be assayed into said diazo solution at a volume ratio of from about 1:100 to about 1:25 to thereby commence an azobilirubin process; and (d) measuring the instantaneous rate of azobilirubin formation at 545 nm in a time range of from about 12 to about 25 seconds after introduction of the aliquot of sample into the diazo solution.

A substrate solution comprising (a) a substrate selected from a group consisting of salts of L- gamma -glutamyl-p-nitroanilide and L- gamma -glutamyl-3-carboxy-p-nitroanilide; and (b) a polyol selected from a group consisting of alkyl polyols containing 2 to 10 carbon atoms and 2 to 10 hydroxy groups and polyethylene glycol having an average molecular weight of from about 200 to about 600. Also, a kit for the determination of gamma -glutamyltransferase activity of the type comprising (a) a buffered medium comprising a buffer and a gamma -glutamyl residue acceptor; and (b) a substrate solution; the buffered medium and substrate solution being adapted such that preselected aliquots thereof can react in the presence of gamma -glutamyltransferase to produce an absorbing cleavage product. The kit is characterized in that the substrate solution is as described above. In, addition, a method for determining gamma -glutamyltransferase activity of the type comprising (a) contacting in any preselected order an aliquot of a buffered medium comprising a buffer and a gamma -glumtamyl residue acceptor, an aliquotof a substrate solution, and an aliquot of a sample to be assayed to thereby form a cleavage product; and (b) measuring the absorbance of said cleavage product. The method is characterized in that the substrate solution is as described above.
Abstract:
An improved motor speed control system for high speed, high resolution motor control applications such as centrifuges. A proportional or proportional plus integral motor control system is provided with improved circuitry which uses digital phase lock techniques to generate a speed difference signal that accurately reflects the difference between the actual and desired speed of the motor. This speed difference signal is used to adjust the speed of the motor until the actual and desired speeds become approximately equal to one another. An error integrating network may then be enabled to eliminate any remaining difference between actual and desired speed, thereby assuring a negligible speed error.
