Abstract: A screening method of lung cancer-specific methylation maker gene is provided to diagnose the lung cancer at the initial transformation stage, thereby accurately and rapidly diagnosing the lung cancer and the progress step of lung cancer. Abstract: A kit and a nucleic acid chip for diagnosing lung cancer are provided to diagnose the lung cancer at an early transformation step, thereby capable of diagnosing the lung cancer at an early stage and diagnosing the lung cancer and the progressing stages of the lung cancer accurately and rapidly. A method for screening a methylated marker gene capable of identifying the transformation of normal cells into lung cancer cells comprises the steps of: (a) comparing a gene expression list of transformed cells with that of non-transformed cells to identify genes with decreased expression from the transformed cell; (b) treating the transformed cells with a demethylating agent and comparing a gene expression list of the transformed cells with that of transformed cells treated with no a demethylating agent to identify genes with more expressed in the demethylating agent-treated cells; and (c) choosing the genes commonly identified from the steps(a) and (b) as a methylated marker gene. A kit for diagnosing lung cancer or lung cancer progressing stage of a body to be tested comprises a primer for amplifying the screened marker gene and an agent for sensitively cleaving a non-methylated nucleic acid selected from the group consisting of MspI, HpaII, BssHII, BstUI and NotI.
