Abstract:
A production method of a transgenic cloned embryo having a goat somatic cell nucleus transfected with hGM-CSF(human granulocyte macrophage-colony stimulating factor) gene is provided to produce a cloned goat expressing the hGM-CSF from the mammary gland, thereby improving the production yield of hGM-CSF. A production method of a transgenic cloned goat embryo comprises the steps of: (1) transforming a somatic cell of goat with a recombinant expression vector containing an hGM-CSF gene to prepare a nucleus donor cell; (2) collecting an ovum from the goat to prepare a nucleus recipient ovum; and (3) denucleating the recipient ovum, transplanting the nucleus donor cell into the recipient ovum and fusing them to prepare the transgenic cloned embryo.
Abstract:
A method for performing animal embryo biopsy using laser is provided to be able to rapidly and conveniently select and transplant only desired fertilized eggs with decreasing damage of the fertilized eggs, thereby increasing production efficiency of domestic animal. The method comprises the steps of: (a) washing fertilized eggs of a cow at the development stage such as 8-cell stage, blastocyst stage and gemmule stage with a buffer solution such as D-PBS and locating it at a buffer solution containing serum; (b) after fixing the fertilized eggs with a compensation pipette where the end portion is inclinedly worked, applying laser having a strength of 40-60 microsec 2-3 times to zone pellucida of the fertilized eggs so as to perforate the zone pellucida; and (c) after locating an enucleation pipette, where the end portion is inclinedly worked, to be opposite to the compensation pipette along the center of the fertilized eggs where the zone pellucida is perforated, applying pressure thereto using two pipettes to emit blastomere.
