본 발명은 줄기세포의 분화유도 방법 및 그로부터 분화된 세포에 관한 것으로, 상세하게는 줄기세포로부터 배상체를 형성하는 단계 및 상기 배상체에 염화리튬을 첨가 배양하는 단계를 포함하는 줄기세포의 분화유도 방법, 상기 방법으로 제조된 세포 및 이를 포함하는 치료용 조성물에 관한 것이다.
Abstract:
A method for inducing differentiation of stem cells is provided to reduce the costs for differentiation by using inexpensive lithium chloride as a differentiation-inducing agent, induce the differentiation phenomenon similar as a normal development process in vitro culture, and increase differentiation efficiency. A method for inducing differentiation of stem cells comprises the steps of: (a) forming embryoid bodies from the stem cells through floating aggregation culture method; and (b) adding 5-20 mM of lithium chloride to the embryoid bodies and culturing them in a culture vessel containing an extracellular substrate such as laminin, collagen or Engelbreth-Holm-Swarm. Further, liver cell separating medicine is added into the embryoid bodies.
Abstract:
PURPOSE: A use of milk fat globule-EGF factor 8(MFGE8) for liver regeneration and liver disease treatment is provided to ensure high expression level of MFGE8 in liver cells which is induced and differentiated in human embryonic stem cells, and to promote proliferation of the liver cells and revascularization. CONSTITUTION: A composition for preventing or treating liver diseases contains MFGE8. The composition further contains a pharmaceutically acceptable carrier. A cell therapeutic agent contains MFGE8 and liver cells. The liver cells are induced and differentiated from human embryonic stem cells.

PURPOSE: A method for transdifferentiation of mesenchymal stem cells into endodermal cells is provided to induce differentiation into endodermal cells including hepatocytes or pancreatic cells. CONSTITUTION: A method for transdifferentiation of mesenchymal stem cells into endodermal cells comprises a step of isolating the mesenchymal stem cells into single cells and forming a sphere. The formation of the sphere comprises a step of adding B27 to a DMEM-low glucose medium. The mesenchymal stem cells are umbilical cord blood-derived mesenchymal stem cells. The medium further contains okadaic acid.

본 발명은 Milk fat globule-EGF factor 8 (MFGE8)을 이용한 간 재생 및 간 질환 개선 용도에 관한 것으로, 보다 상세하게는 MFGE8는 인간 배아줄기세포에서 유도 분화된 간세포에서 높은 발현율을 나타내며, 간세포 증식 및 혈관 재생을 촉진하여 단독으로 사용하거나, 간세포와 함께 사용하여 간 재생 및 간 질환 개선을 달성할 수 있다.

본 발명은 중간엽 줄기세포를 단일세포(single cell)로 분리하고, 분리한 단일세포 집단으로부터 구체(sphere)를 형성하는 단계를 포함하는, 중간엽 줄기세포에서 내배엽성 세포로의 분화를 유도하는 교차 분화 방법을 제공한다. 상기한 본 발명에 의하면, 중배엽성 세포인 다양한 중간엽 줄기세포들로부터 완벽한 기능을 하는 간세포 또는 췌장세포를 포함한 내배엽성 세포로의 분화를 유도할 수 있는 이점이 있다.

PURPOSE: A composition containing a conditioned medium which is prepared by culturing liver cells from human embryonic stem cells is provided. CONSTITUTION: A composition for treating liver diseases contains a conditioned medium which is prepared by dyeing ICG(indocyanine green) among cell differentiated from human embryonic stem cells and culturing ICG+ liver cell colony. An animal cell medium contains DMEM/F12, insulin, transferring, and selenium.
