Abstract: PURPOSE: A method for preparing embryonic stem cell-like cells from somatic cells is provided to evaluate chemical or drug toxicity. CONSTITUTION: A method for preparing embryonic stem cell-like cells from somatic cells comprises: a step of culturing fibroblasts in a medium; a step of infecting with packaging cells by performing transfection of vector which is treated with oxysterol and contains Oct4 gene to the fibroblasts; and a step of culturing the infected fibrobasts.

Abstract: 본 발명은 a) 옥시스테롤 (Oxysterol); 및 b) Oct4 단백질 또는 Oct4 단백질을 코딩하는 핵산분자를 포함하는 체세포로부터 배아줄기세포 유사세포로의 역분화를 유도하는 조성물 및 이를 이용한 배아줄기세포 유사세포의 제조방법에 관한 것이다. 또한, 본 발명은 Bmi1의 상위 조절인자인 Shh 유사체(analog)인 옥시스테롤 (oxysterol; hydroxycholesterol)을 처리하여 신경줄기세포와 같은 세포로의 유도를 하면서 Oct4 유전자를 도입하여 배아줄기세포 유사세포를 제조함으로써 역분화를 위한 도입 유전자 수를 감소시킬 수 있는 새로운 역분화 유도 방법을 제시하고, 본원발명에 의해 제조된 배아줄기세포 유사세포는 배아줄기세포와 같이 전분화능을 가져 각종 질병치료 등에 유용하게 이용될 수 있다.

Abstract: PURPOSE: A composition for inducing redifferentiation from somatic cells to embryonic stem cell-like cells is provided to treat various diseases. CONSTITUTION: A composition contains purmorphamine; and a virus which expresses Oct4 protein by introduction of a nucleic acid molecule encoding Oct4 protein. A method for preparing embryonic stem cell-like cells comprises: a step of culturing fibroblast in a medium; a step of performing transfection of a vector to the cultured fibroblasts and infecting packaging cells; and a step of culturing the fibroblasts.
