Abstract: 본발명은 Atg5 유전자를과발현하여향상된기저자식작용에의해수명이연장된형질전환동물, 및이의제조방법에관한것이다. 본발명에따른 Atg5 유전자형질전환동물은평균수명이 WT 마우스보다더 연장되며, Atg5 형질전환마우스의 MEFs에서 Atg5에의해유도된향상된자식작용활성을나타내고, 이러한향상된자식작용활성이산화적스트레스에의해유도된세포사멸에대한내성에중요하며, Atg5 형질전환마우스의 MEFs에서산 β-갈락토시다제활성이 WT 마우스의 MEFs에비해감소한다. 또한, 본발명에따른 Atg5 형질전환동물은비만억제에탁월한효과가있다. 따라서, 본발명에따른 Atg5 형질전환동물은 Atg5 유전자를과발현하여향상된기저자식작용에의해수명을연장시킬수 있음으로, Atg5 유전자형질전환동물은수명연장동물모델로유용하게이용할수 있다.

Abstract: PURPOSE: A lifespan-extended transgenic animal and a method for preparing the same are provided to extend the life span of an animal by overexpressing Atg5 gene. CONSTITUTION: A method for preparing a lifespan-extended transgenic animal is as follows. After Atg5 gene is sub-cloned in a transformation vector, a transformation cassette is separated. The separated transformation cassette is injected into the mammal embryo bud except for the human and fertilized egg is manufactured. The transformation animal is produced by implanting a fertilized egg in the surrogate mother mammal except for the human.

Abstract translation: 目的：提供延长寿命的转基因动物及其制备方法，以通过过表达Atg5基因延长动物的寿命。 构成：用于制备延长寿命的转基因动物的方法如下。 将Atg5基因亚克隆到转化载体中后，分离转化盒。 将分离的转化盒注射到哺乳动物胚芽中，除了人和受精卵被制造。 转化动物通过将受精卵植入代孕母亲哺乳动物而不是人来产生。
