Abstract: 본 발명은 파골세포형성을 저해하는 폴리펩티드, 그를 포함하는 약학적 또는 식품 조성물, 그를 이용한 개체의 파골세포형성을 저해하는 방법 및 골질환을 치료 또는 예방하는 방법, 그를 코딩하는 핵산, 상기 핵산을 포함하는 벡터 및 숙주세포에 관한 것이다. 본 발명의 파골세포형성을 저해하는 폴리펩티드는, 기존의 알려진 RANKL 신호 전달에는 전혀 영향을 주지 않고, RANK와 RANKL의 상호작용보다 하류에서의 신호전달을 저해하여, 파골세포의 기능에 필요하다고 알려진 액틴 고리의 형성에 관련된 경로를 차단함으로써 파골세포의 형성을 저해하므로, 종래 알려진 약물에 비하여 더 적은 부작용을 갖는다. 본 발명의 약학적 또는 식품 조성물에 의하면, 개체의 파골세포형성 (osteoclastogenesis)을 저해하여 골질환을 치료 또는 예방하기 위한 용도로 사용될 수 있다. 본 발명의 개체의 파골세포형성 (osteoclastogenesis)을 저해하는 방법에 의하면, 개체의 파골세포형성을 효과적으로 억제하여, 골질환을 치료 또는 예방할 수 있다.

Abstract: A polypeptide inhibiting RANK(receptor activator of NF(nuclear factor)-kappaB) is provided to inhibit osteoclast differentiation by RANKL(RANK ligand), bone destruction function by activated osteoclast, cell fusion of osteoclast precursor and ring formation of actin protein in osteoclast, so that the polypeptide is useful for preventing and treating diseases caused by differentiation or activation of osteoclast. A polypeptide inhibiting RANK consists of 6-12 consecutive amino acids selected from amino acids contained in the amino acid sequence of SEQ ID NO:1, wherein the polypeptide contains 4th-9th amino acids in the amino acid sequence of SEQ ID NO:1, and is linked to a cell permeable polypeptide as a leader signal sequence in the N-terminal or C-terminal. A pharmaceutical composition for preventing and treating a disease caused by differentiation or activation of osteoclast selected from osteoporosis, arthritis, bone destruction caused by cancer, leukemia and multiple myeloma comprises the polypeptide inhibiting RANK and pharmaceutically acceptable carriers.

Abstract: PURPOSE: A polypeptide which suppresses osteoclast formation is provided to suppress signal transduction of downstream of RANK and RANKL(receptor activator of nuclear factor-kB ligand) and prevent or treat bone diseases without side effects. CONSTITUTION: A peptide which suppresses osteoclast formation is denoted by general formula Ile-Ile-Xaa_1-Xaa_2-Xaa_3-Val. The Xaa_1 and Xaa_2 are independently Ile or Val. The polypeptide is a polypeptide of sequence number 33, 44, 55, 56, 65, 80, 81, 84, 85, or 86. A pharmaceutical composition for suppressing osteoclastogenesis of an individual contains the polypeptide and pharmaceutically acceptable carrier. Bone diseases are prevented or treated by administering the pharmaceutical composition to an individual. The bone diseases include arthritis, osteoporosis, Paget's disease, metabolic bone disease, leukemia or multiple myeloma.
