    Abstract: A DNA vaccine comprising a gene encoding HPV(human papillomavirus) protein is provided to treat or prevent cervical cancer by effectively inducing humoral immune response and cellular immune response, and increase immune response-inducing efficiency with a relatively small amount of antigen within a short time by injecting the vaccine into dermis by using a tattoo device. A DNA vaccine for preventing and treating cervical cancer comprises an E5 gene of SEQ ID NO:14 and L1 gene of SEQ ID NO:10 and/or L2 gene of SEQ ID NO:12 of HPV selected from HPV genotypes 16, 18, 26, 30, 31, 33, 34, 35, 39, 45, 51, 52, 53, 56, 58, 59, 61, 66, 67, 69, 70 and 73, wherein the E5, L1 and L2 gene are contained in a separate plasmid. A vaccine composition comprises the DNA vaccine and pharmaceutically acceptable carriers or immunoadjuvant selected from LTB(leukotriene B), IL-15(interleukin-15) and GM-CSF(Granulocyte macrophage-colony stimulating factor), and is administered into the dermis or muscle through a tattoo device. A cell line KCLRF-BP-00140 stably expresses the E5 gene of HPV and is used for screening a material, vaccine or drug for diagnosis or treatment of HPV infection.
