Abstract: PURPOSE: An APPL1(Adaptor protein, Phosphotyrosine interaction, PH domain and Leucine zipper containing 1) inhibitor is provided to reduce EGFR(epidermal growth factor receptor) expression and to treat cancer caused by EFGR overexpression. CONSTITUTION: A composition for preventing and treating cancer contains a material which suppresses expression or activation of APPL1. The material is siRNA(small interfering RNA), shRNA(short hairpin RNA) or antisense nucleic acid. The siRNA has a sequence of sequence number 1 or 2. The material which suppresses APPL1 activation is an antibody to BAR domain or PH domain. A method for screening a therapeutic agent for cancer comprises: a step 1 of checking binding amount of APPL1 and EGFR in cells of a patient with cancer; a step 2 of treating with a candidate; a step 3 of confirming the binding amount; and a step 4 of comparing the expression level of steps 1 and 3.

Abstract translation: 目的：提供APPL1（适配蛋白，磷酸酪氨酸相互作用，PH结构域和亮氨酸拉链含有1）抑制剂，以减少EGFR（表皮生长因子受体）表达并治疗由EFGR过表达引起的癌症。 构成：用于预防和治疗癌症的组合物含有抑制APPL1的表达或活化的材料。 该材料是siRNA（小干扰RNA），shRNA（短发夹RNA）或反义核酸。 siRNA具有序列号1或2的序列。抑制APPL1活化的物质是对BAR结构域或PH结构域的抗体。 筛选癌症治疗剂的方法包括：检查患有癌症的患者的细胞中APPL1和EGFR的结合量的步骤1; 与候选人对待的第二步; 确认结合量的步骤3; 以及比较步骤1和3的表达水平的步骤4。
