Abstract: 본 발명은 Mis18α 유전자 넉아웃 생쥐 및 그 배아에 관한 것으로서, 보다 상세하게는 Mis18α 유전자 일부와 조건적 넉아웃(conditional knockout) 생쥐모델의 제작에 사용되는 서열들을 포함하는 적중벡터(targeting vector), 상기 적중벡터를 이용하여 생성된 배아간세포(Embryonic Stem Cell), 이를 이용하여 얻은 Mis18α 넉아웃 생쥐 배아, 그 제조방법 및 이용방법 등에 관한 것이다. Mis18α는 배아세포 분열시 염색체 분열과정에서 중요한 역할을 수행하며 특히 배아 세포의 발달과정에 있어서 매우 중요한 인자가 될 수 있다. 따라서 본 발명의 Mis18α 넉아웃 생쥐모델은 염색체 분열과정이 생체에 미치는 영향과 그 이상으로 암이 발생하는 기작 등을 밝힐 수 있는 좋은 실험모델이 될 수 있으며, 신약개발의 표적이 될 수 있는 염색체 분열과정에 관여하는 신규한 단백질들을 동정하는데 활용될 수 있다. 나아가 동원체의 형성과정에 관여하는 다수의 신규 유전자들을 확보하고, 동원체 형성과정에 이상이 생겼을 경우 나타날 수 있는 질환을 분석하는 기술을 개발하는데 이용될 수 있다.

Abstract: PURPOSE: A Mis 18alpha gene knockout mouse and a method for producing the same are provided to identify effects of chromosome division process to a body and carcinogenic mechanism. CONSTITUTION: A mouse Mis 18alpha targeting vector comprises: LoxP (locus of X-over P1) site; a gene fragment from EcoRI cleavage site which is secondarily adjacent to 5' of a first exon to a second exon; neomycin resistant gene; and the EcoRI cleavage site. A method for producing a Mis 18alpha gene knockout mouse comprises: a step of preparing the mouse Mis 18alpha gene targeting vector; a step of introducing the vector into an embryonic stem cell and obtaining an embryonic stem cell clone in which a part of Mis 18alpha gene is deleted; a step of microinjecting the Mis 18alpha knockout embryonic stem cells into an inner cell mass of blastocyte of a normal mouse and implanting on a surrogate mother mouse to obtain a chimera mouse; and a step of hybridizing the chimera mouse and obtaining a Mis 18alpha gene knockout mouse with Mis18α^+/delta genotype.

Abstract translation: 目的：提供错误的18α基因敲除小鼠及其制备方法，以鉴别染色体分裂过程对人体和致癌机制的影响。 构成：小鼠Mis18α靶向载体包含：LoxP（X-over P1位点）; EcoRI切割位点的第二个外显子的第二外显子5'的第二个邻接的基因片段; 新霉素抗性基因; 和EcoRI切割位点。 一种制备Mis18alpha基因敲除小鼠的方法包括：制备小鼠Mis18alpha基因靶向载体的步骤; 将载体引入胚胎干细胞并获得缺失了一部分Mis 18alpha基因的胚胎干细胞克隆的步骤; 将Mis 18alpha敲除胚胎干细胞显微注射入正常小鼠的胚细胞的内细胞块并植入代孕母鼠以获得嵌合体小鼠的步骤; 以及与嵌合体小鼠杂交并获得具有Mis18α^ + /δ基因型的Mis18alpha基因敲除小鼠的步骤。
