Abstract: 본 발명은 치매 (알츠하이머 병) 질환의 진단, 예방 및 치료제 그리고 이들의 스크리닝 방법에 관한 것으로, 구체적으로는 FcγRⅡb와 그의 변이체, FcγRⅡb 세포외 도메인 단백질, 항-FcγRⅡb 항체, FcγRⅡb 특이 펩티드, 및 FcγRⅡb 특이 siRNA로 이루어진 군으로부터 선택되어진 결합 억제제는 FcγRⅡb와 Aβ의 결합을 저해하여 Aβ에 의한 신호 전달, 세포내 이동, 신경 독성, 세포 사멸도 및 기억력 감퇴를 감소시키므로 알츠하이머 질환의 진단, 예방 및 치료에 유용하게 이용될 수 있다.

Abstract: Disclosed herein are methods of diagnosing, preventing and treating Alzheimer's disease based on the use of an inhibitor for the binding of amyloid-&bgr; (A&bgr;) to FcγRIIb, and a method of screening the inhibitor. The inhibitor is selected from the group consisting of an FcγRIIb protein or a variant thereof, an FcγRIIb extracellular domain, an anti-FcγRIIb antibody, an FcγRIIb-specific peptide and an FcγRIIb-specific siRNA. The inhibitor reduces the toxic signaling and intracellular translocation of A&bgr; and the neurotoxicity, neuronal cell death and memory impairment mediated by A&bgr; by inhibiting the binding between A&bgr; and FcγRIIb. Thus, the inhibitor is useful in the diagnosis, prevention and treatment of Alzheimer's disease.
