Abstract:
본 발명은 대장균에서 발현된 내포체 (inclusion body)로부터 재조합 Bax 억제제-1 (BI-1)의 동시 리폴딩 및 재구성에 관한 것이다. 기능적 분석은 결과적으로 수득되는 프로테오리포좀 (proteoliposome)이 산성 환경에 반응하고 리포좀으로부터 포접된 Ca 2+ 의 방출을 유발한다는 것을 보여주었다. 원편광 이색성 분광분석 (Circular Dichoism)에 의해 결정된, 재구성된 BI-1의 이차 구조는 산성 환경에 노출시 α-헬릭스 함량의 증가 및 랜덤 구조의 감소를 입증했다. 이 구조적 변화가 BI-1의 양성자-유도 Ca 2+ 방출을 초래하는 것일 수 있다. PURPOSE: A peptide antigen with enhanced immunogenicity and a method for producing the same are provided to enhance immunogenicity and to increase antibody production efficiency. CONSTITUTION: A peptide antigen with enhanced immunogenicity comprises an amino acid sequence with two or more tandem repeat amino acid sequence unit. The peptide antigen additionally contains proline between amino acid sequence units. A vaccine composition contains the peptide antigen, nucleic acid encoding the same, or antibody to the peptide antigen. A method for producing the peptide antigen with immunogenicity comprises: a step of determining amino acid sequence corresponding to an epitope of a target antigen; and a step of preparing a polypeptide comprising two or more tandem repeat amino acid sequence units; or a step of preparing amino acid sequence which additionally contains proline.

PURPOSE: A method for refolding and recombining recombinant Bas inhibitor-1(BI-1) is provided to use for in vitro functional analysis of BI-1 and to induce Ca^2+ release. CONSTITUTION: A method for producing a liposome containing BI-1 protein comprises: a step of separating an inclusion body containing BI-1 protein; a step of dissolving charotropic agent of first concentration, reductant, and amphoteric surfactant in a first buffer; a step of mixing the first buffer with phospholipid; and a step of exchanging the first buffer to a second buffer. The charotropic agent is urea or guanidiniumchloride.

PURPOSE: A method for preparing a peptide antigen with enhanced immunogenicity is provided to enhance antibody-producing efficiency. CONSTITUTION: A polypeptide contains an amino acid sequence with two or more tandem repeat LEDTQKH amino active sequences and additionally contains one or two praline between the repeat amino acid sequences. The polypeptide is linked to polystyrene or polylactic-glycolic acid polymers or bovine serum albumin(BSA) or ovalbumin. A vaccine composition contains: the polypeptide with two or more tandem repeat LEDTQKH amino active sequences; the polypeptide with LEDTQKH sequences and proline; the polypeptide linked by the polystyrene or polylactic-glycolic acid polymers or bovine serum albumin(BSA) or ovalbumin; a nucleic acid encoding one of polypeptides; and an antibody for one of the polypeptides.
