    Abstract: PURPOSE: A nanocomposite for reconstruction or regeneration of chondrocytes and a method for differentiating stem cells into chondrocytes are provided to effectively deliver hSOX9 gene and siRNA to mesenchymal stem cells. CONSTITUTION: A nanocomposite for reconstruction or regeneration of chondrocytes is prepared by binding hSOX9 gene or nucleotide siRNA containing a base sequence of sequence numbers 1-6 on nanoparticles of biodegradable polymers. The biodegradable polymers are a poly(lactic-co-glycolic acid). The weight ratio of the hSOX9 gene and biodegradable polymers is 1:40-100. A method for differentiating mesenchymal stem cells into chondrocytes comprises a step of culturing the mesenchymal stem cells under the presence of the nanocomosite. A pharmaceutical composition for reconstruction or regeneration of chondrocytes contains the nanocomposite-introduced mesenchymal stem cells and pharmaceutically acceptable carrier.
