Abstract:
본 발명은 병원성 미생물 탐지용 프로브 및 이를 이용한 살아 있는 병원성 미생물 검출방법에 관한 것으로, 구체적으로 본 발명은 기존의 표지 방식과는 달리 표지 물질을 포함하고 있는 ribosomal RNA의 탐지용 프로브의 개발과 이를 이용하여 임상 샘플 속의 살아 있는 병원성 미생물을 매우 신속하고 정확하게 검출할 수 있는 방법이다.
Abstract:
PURPOSE: A probe for detecting CarB gene of Salmonella is provided to enhance specificity compared with an existing probe for detecting 16S rDNA, thereby detecting and distinguishing Salmonella serotypes. CONSTITUTION: A probe composition for detecting Salmonella contains a polynucleotide having a base sequence with 15-25 continuous bases in CarB gene of Salmonella or a complementary base sequence thereof. The polynucleotide is selected among polynucleotides with base sequences of sequence numbers 1-7 or complementary base sequences thereof and is used for detecting Salmonella enterica. The polynucleotide is selected among polynucleotides with base sequences of sequence numbers 2-7 or complementary base sequences thereof and is used for detecting Salmonella serotypes. PURPOSE: A probe for detecting ribosomal RNA and a method for detecting pathogenic microorganism using the same are provided to ensure quick and accurate detection. CONSTITUTION: A method for detecting microbes comprises: a step of preparing cell lysate containing ribosomal RNA from a test sample; a step of mixing a detecting probe and capturing probe in the cell lysate for hybridization; and a step of confirming hybridization. The detecting probe is labeled with a marker at 5' terminal or 3' terminal. The detecting probe contains a base sequence of sequence number 28. The capturing probe contains a base sequence of sequence number 6. The microbe is E. coli O157:H7. A probe for detecting Listeria monocytogenes contains a base sequence of sequence number 24.

PURPOSE: A carbohydrate chip for detecting vibrio cholera is provided to detect small amount of cholera toxin generated from vibrio cholera and detect cholera toxin at the same time by using small amount of sample based on the chip. CONSTITUTION: A carbohydrate chip for detecting vibrio cholera comprises a solid substrate of which surface is comprised of GM1 pentasaccharide, GM2 tetrasaccharide, asialo GM1 tetrasaccharide, GM3 trisaccharide, Galactose-β1,3 N-acetylgalactosamin, lactose and sialic acid. The carbohydrate chip introduces (-NH 2) group GM1 pentasaccharide, GM2 tetrasaccharide, GM3 trisaccharide, GM3 trisaccharide, Galactose-β1,3 N-acetylgalactosamin, lactose and sialic acid with sialic acid amine group. The solid substrate is introduced to more than one functional group chosen from aldehyde, ketone, N-hydroxy-succinimide, epoxide, imido- ester, anhydride in the solid substrate and carbonate. GM1 pentasaccharide, GM2 tetrasaccharide, asialo GM1 tetrasaccharide, GM3 trisaccharide, Galactose-β1,3 N-acetylgalactosamin, lactose and sialic acid is fixed to the solid substrate.
