Abstract: 본 발명은 엉겅퀴 추출물을 유효성분으로 하는 골다공증 치료 및 예방용 조성물에 관한 것으로 상기 엉겅퀴 추출물은 엉겅퀴를 분쇄한 후, 상기 분쇄된 엉겅퀴와 주정을 혼합하여 제조한 엉겅퀴 추출물을 유효성분으로 하는 골다공증 치료 및 예방용 조성물에 대한 것이다.
또한, 상기 엉겅퀴 추출물은 조골전구세포에서 조골세포로 분화하는 과정에서 염기성 인산분해효소의 활성을 증가시키는 것은 물론이며, 대식세포의 파골세포 분화과정에서 골형성을 촉진시키고 골흡수를 억제시키는 효능이 있다.
또한, 상기 엉겅퀴 추출물과 이소플라본을 혼합하였을 경우 골다공증 예방과 치료에 있어서 이소플라본과의 상승 효능을 발휘할 수 있다.
상기의 방법으로 제조된 엉겅퀴 추출물을 유효성분으로 하는 골다공증 치료 및 예방용 조성물은 골형성을 촉진시키고 골흡수를 억제시켜 주는 효능으로 골다공증을 치료하고 예방할 수 있다.

Abstract: 본 발명은 골형성을 도모하는 조골세포 분화를 촉진시키고 골흡수를 도모하는 파골세포의 골흡수능을 억제시키는 실리비닌(silibinin)을 포함하는 골다공증 치료 및 예방용 조성물에 대한 것이다. 보다 구체적으로, 실리비닌은 골형성에 관여하는 조골전구세포(마우스 섬유아세포인 MC3T3-E1)의 분화 단계 중 염기성 인산분해효소(alkaline phosphatase)의 활성을 증가시키고, 제 1형 콜라겐(type 1 collagen) 생성을 촉진시킴으로서 조골세포의 석회화를 촉진시킨다. 또한 대식세포(마우스 대식세포인 RAW264.7)를 파골세포로 분화시킨 후 파골세포의 골흡수 과정에서 기질단백질 분해효소(matrix metalloproteinase, MMP)-9 활성을 억제시킨다. 이로서 실리비닌은 부작용이 없으면서 조골세포의 석회화를 촉진시키며, 파골세포의 골흡수를 억제시킬 수 있는 골다공증 치료 및 예방할 수 있는 조성물에 대한 것이다.

Abstract: The present invention relates to a composition for treating and preventing osteoporosis which includes phloretin which suppresses bone resorption of the osteoclast which promotes bone resorption due to osteoporosis. More specifically, the present invention relates to the composition for treating and preventing osteoporosis in which phloretin reduces the expression of carbonic anhydrase and V-ATPase D2 which participate in acidification condition during bone resorption of the osteoclast and suppresses secretion of MMP-9 secreted during bone resorption and suppresses bone resorption of the osteoclast by reducing the expression and secretion of Cathepsin K. In addition, bone density, which is reduced by ovariectomization in an ovariectomized animal model, is increased. Therefore, phloretin suppresses the differentiation of the osteoclast without side effects and suppresses bone resorption.

The extract is prepared by mixing yellow flag iris with water, alcohol, acetone, hexane, ethyl acetate, or a mixture thereof, heating at 60-70 deg.C., filtering and concentrating the extract, and freeze-drying the extract. The composition enhances alkaline phosphatase activity and promotes calcification.
