CONSTITUTION:Yeast such as Saccharomyces cerevisiae, etc., belonging to the genus Saccharomyces, capable of producing riboflavin, is inoculated into a medium containing a carbon source such as glucose, acetic acid, ethanol, etc., a nitrogen source such as ammonium sulfate, peptone, urea, etc., and a phosphoric acid source such as phosphate, etc. The culture is carried under an aerobic condition out by continuously and intermittently supplying the consumed carbon source, nitrogen source and phosphate source. Riboflavin is collected from the prepared culture solution by a well-known method.

    Abstract: PURPOSE:To efficiently obtain the titled enzyme useful for producing a blended amino acid substance, etc., by cultivating a specific bacterium belonging to the genus Pseudomonas or Trichosporon in a nutrient culture medium. CONSTITUTION:Pseudomonas.fluorescens IFO12055 or Trichosporon sp. 107-1 (FERM P-9489) is cultivated in a culture medium containing 0.5-2% (wt./vol.) carbon source, 0.3-1% (wt./vol.) nitrogen source, 0.05-0.1% (wt./vol.) magnesium, 0.001-0.003% (wt./vol.) calcium, manganese and ferric ion and 0.5-1% (wt./vol.) glycyl-L-proline, polypeptone, etc., at pH5-8 and 20-40 deg.C for 18-40hr. Microbial cells are then collected from the resultant culture and crushed to isolate the aimed proline dipeptidase from the obtained crude extract solution.

    Abstract: PURPOSE:To easily obtain D-lactic acid having high optical purify, by culturing a microbial strain assimilating L-lactic acid in a medium containing racemic lactic acid or a mixture of the recemic compound and an optically active lactic acid or by contacting the racemic lactic acid, etc., with cultured microorganism. CONSTITUTION:A culture medium is prepared by using racemic lactic acid or a mixture of racemic lactic acid and an optically active lactic acid as a carbon source and compounding the source with a nitrogen source such as urea, inorganic salt such as MgSO4 and, if necessary, a proliferation promoting factor such as yeast extract. The obtained medium is inoculated with a microbial strain (e.g. Bacillus subtilis IFO 3134) belonging to Bacillus genus and having an assimilation rate of L-lactic acid high than that of D-lactic acid. The strain is cultured in the medium at 3-10pH and 20-60 deg.C for several hours several days under supply of air. The microbial cells are separated and removed from the cultured product and the supernatant liquid is concentrated and refined by ion exchange treatment, solvent extraction, etc., to obtain the objective D-lactic acid.

    Abstract: PURPOSE:To attain high bacterial cell yield, by culturing obligate anaerobe at high concentration in a liquid medium added with water-extracted or dialyzed agar. CONSTITUTION:Water is added to agar, maintained in suspended state and centrifuged to obtain an extract. As an alternative method, agar wet with water or suspended in water is put into a tubular dialysis membrane and dialyzed in water to obtain a dialyzed liquid. The extract or dialyzate is added to a conventional liquid medium at a rate of about 1-30wt% in terms of agar used in the extraction of dialysis. An obligate anaerobe such as methanol-assimilating bacteria, etc., is cultured in the liquid medium.

    Abstract: PURPOSE:To enable stable production of vitamin B12 at a low cost suppressing the dissipation of methanol, by carrying out anaerobic culture of a specific methanol-assimilating microorganism in a medium containing methanol as a main carbon source. CONSTITUTION:A microbial strain belonging to Acetobacterium genus such as Acetobacterium woodii DSM1030 is inoculated to a medium containing methanol as a main carbon source, a nitrogen source such as urea, inorganic salts such as Co salt and, if necessary, yeast extract, 5,6-dimethylbenzimidazole (a vitamin B12 precursor), etc., and cultured under anaerobic condition in a gaseous phase composed of N2, CO2, etc., at 20-40 deg.C and 5.5-8.5pH for 3-10 days without or under shaking. Vitamin B12 produced and accumulated in the culture product is separated by phenol extraction, column chromatography, etc.
