Abstract: A hybrid plasmid with the replicon intrinsic to the methylotrophic bacterium is obtained from a plasmid from obligately methylotrophic bacteria, e.g. Methylomonas, on the one hand and a plasmid with selection markers on the other hand. This hybrid plasmid is - where appropriate after previous insertion of further genetic information - introduced by transformation into a host organism such as E. coli. After amplification and appropriate selection, the clones are treated with a suitable conjugative plasmid, and the mobilisability defect is eliminated by introducing another suitable plasmid. The clones obtained in this way are conjugated with plasmid-free obligately methylotrophic bateria as recipient, resulting - after selection - in clones which express proteins in accordance with the inserted genetic information.
