Abstract: PROBLEM TO BE SOLVED: To provide a DNA which contains a promoter of period 1 gene and can be used for elucidating the mechanism of the expression regulation of period 1 gene, and a system and a method for observing an in vivo expression amount of a target gene. SOLUTION: This DNA has two first exons of period 1 gene that are selectively transcripted and two promoter regions having a function as a promoter of the period 1 gene. A non-human animal having the DNA has a DNA construct that is integrated upstream of a reporter gene. The system/method for observation comprises the steps of inserting an optical fiber into an observation region of the non-human animal, obtaining a luminescence generated there, and quantifying it by using a photon counter.
