Abstract:
Analyzers and analyzer systems that include an analyzer for determining multiple label species, methods of using the analyzer and analyzer systems to analyze samples, are disclosed herein. The analyzer includes one or more sources of electromagnetic radiation to provide electromagnetic radiation at wavelengths within the excitation bands of one or more fluorophore species to an interrogation space that is translated through the sample to detect the presence or absence of molecules of different target analytes. The analyzer may also include one or more detectors configured to detect electromagnetic radiation emitted from the one or more fluorophore species. The analyzer for determining multiple target molecule species provided herein is useful for diagnostics because the concentration of multiple species of target molecules may be determined in a single sample and with a single system.

Cardiovascular disease, e.g., congestive heart failure, is often first diagnosed after the onset of clinical symptoms, eliminating potential for early intervention. The invention provides a multi-marker immunoassay, including cardiac pathology and vascular inflammation biomarkers, yielding a more sensitive assay for early detection of CHF in plasma. A panel consisting of cardiac pathology (cTnl, BNP) and vascular inflammation (IL-6, TNFα, IL- 17a) biomarkers provided a sensitivity of 94% for association with CHF.
Abstract:
Disclosed are methods, kits, and compositions for fluorescent detection of single biomolecules, e.g. proteins, using highly sensitive fluorescent dyes and a flow cell with one interrogation space for irradiation by focused laser beam. The methods, kits, and compositions are useful in determining concentrations of molecules in samples to levels of 1 femtomolar, 1 attomolar, or lower. The methods, kits, and compositions also allow the determination of concentration over a wide range, e.g., 7-log range, without need for sample dilution.
Abstract:
A detector captures an image in response to one or more optical elements directing a first electromagnetic radiation to a binding surface. The image is defined by a plurality of pixels. Each pixel provides a signal. The signals provided by a subset of the pixels results at least in part from a second electromagnetic radiation emitted by target molecules bound to the binding surface, in response to the first electromagnetic radiation. The subset of pixels indicates a presence of the target molecules in corresponding areas of the binding surface. A processor determines a noise signal threshold based on an average background noise signal for the plurality of pixels. The processor identifies the subset of pixels indicating the presence of the target molecules by identifying the pixels having a signal greater than the noise signal threshold. The processor determines a count of the target molecules based on the subset of pixels.

The disclosure is directed to a system including a microfluidic cartridge having a fluidic pathway and onboard reagents for processing a sample for analysis. The system includes an an optics module including an electromagnetic radiation source, an objective, and a detector, and a translating that translates at least one of the cartridge or the optics module so as to scan a processing sample with focused electromagnetic radiation. Analysis methods using the system are disclosed.
